基于拉曼光谱的家蚕卵滞育解除无损检测

摘  要:家蚕是以卵态滞育的昆虫,其滞育性与养蚕生产密切相关。以大造和9·芙×7·湘两品种的滞育和解除滞育蚕卵为样本,使用拉曼光谱仪采集各样本的光谱数据,对光谱数据使用三次样条插值(Spline)、自适应迭代惩罚最小二乘法(airPLS)等预处理操作,分别构建偏最小二乘判别分析(partial least squares discriminant analysis,PLSDA)模型和支持向量机(support vector machine,SVM)模型。结果表明,大造蚕卵PLSDA模型的准确率达0.987,AUC(area under curve)值为0.975;9·芙×7·湘蚕卵PLSDA模型准确率达0.975,AUC值为0.999;而两品种混合蚕卵的PLSDA模型准确率达0.975,AUC值为0.999。大造、9·芙×7·湘和两品种混合蚕卵的SVM模型准确率分别为0.925、0.950、0.944,AUC值分别为0.943、0.962、0.964。PLSDA建模效果略好于SVM。此外,根据拉曼峰面积大小、位置和PLSDA回归系数矩阵3种方法提取蚕卵拉曼光谱特征峰,实现对家蚕卵滞育性区分,准确率最高达0.963。综上所述,基于拉曼光谱的家蚕卵滞育解除无损检测方法具有良好的效果,为家蚕卵滞育性的快速检测提供了新途径。Bombyx mori is a diapause insect in the form of egg,and its diapause is closely related to sericulture production.In this study,the spectral data of the diapause and diapause termination silkworm eggs of Dazao and 9·Fu×7·Xiang were collected by Raman spectroscopy and preprocessed by Spline interpolation and airPLS.Two models,namely partial least squares discriminant analysis(PLSDA)and support vector machine(SVM),were constructed respectively.The results showed that the two models had good prediction effect.For samples of Dazao Bombyx mori eggs,the accuracy of PLSDA model was 0.987,and the value of AUC(area under curve)was 0.975.For 9·Fu×7·Xiang,the accuracy of PLSDA model was 0.975,and the value of AUC was 0.999.For the mixed eggs of two varieties,the accuracy of the PLSDA model was 0.975,and the value of AUC was 0.999.For the SVM model constructed by Dazao,9·Fu×7·Xiang and the mixed eggs of two varieties,the accuracy was 0.925,0.950 and 0.944 respectively,and the value of AUC was 0.943,0.962 and 0.964 respectively.The modeling effect of PLSDA is slightly better than that of the SVM.In addition,according to the size and location of Raman peak area and PLSDA regression coefficient matrix,the characteristic peaks of Raman spectrum of Bombyx mori eggs were extracted to distinguish the diapause states of Bombyx mori eggs with an ideal accuracy as high as 0.963.In conclusion,the nondestructive testing method of diapause termination of Bombyx mori eggs based on Raman spectroscopy has good effect,which provides a new way for rapid detection of diapause state of Bombyx mori eggs.
