摘 要:目的观察薯蓣皂苷对甲状腺癌细胞SW579细胞的抑制作用并探讨其可能的作用机制。方法体外常规培养甲状腺癌SW579细胞,将细胞分为对照组、不同浓度薯蓣皂苷组(10、20、30、40、50μM),CCK-8方法检测各组甲状腺癌SW579细胞存活率;随后将细胞分为对照组、薯蓣皂苷(30μM)组、Jagged 1组(5mg/L)组及薯蓣皂苷+Jagged 1组。EDU染色观察各组细胞增殖情况;流式细胞仪检测细胞凋亡率;Real-time PCR检测凋亡基因Bax、 Bcl-2及Caspase-3基因表达;Transwell实验观察各组细胞侵袭能力;Western blot检测Notch 1、Jagged 1及Hes 1蛋白表达。结果薯蓣皂苷能抑制SW579细胞活性,抑制其增殖;能够促进SW579细胞凋亡,增加Bax/Bcl-2比例,增加Caspase-3基因表达;并抑制细胞侵袭;薯蓣皂苷还可下调Notch1、Jagged 1及Hes 1蛋白表达(P<0.05);并且薯蓣皂苷部分逆转了Jagged 1的作用。结论薯蓣皂苷对甲状腺癌细胞SW579细胞具有一定的抑制作用,其作用机制可能在一定程度上与抑制Notch 1信号通路有关。Objective To observe the inhibitory effect of dioscin on thyroid carcinoma SW579 cells and explore its possible mechanism. Methods Thyroid carcinoma SW579 cells were cultured in vitro according to the conventional method, and the cells were divided into control group and dioscin groups with the series of concentrations(10, 20, 30, 40, 50 μM). The survival rate of thyroid carcinoma SW579 cells were detected by CCK-8 method. The cells were divided into control group, dioscin(30 μm) group, Jagged group 1(5 mg/L) group and dioscin+Jagged 1 group. Cell proliferation was observed by EDU staining. Cell apoptosis rate was detected by flow cytometry. The expression of Bax, Bcl-2 and Caspase-3 mRNA was detected by Real-time PCR. Cell invasion ability was observed by Transwell. Notch 1, Jagged 1 and Hes 1 protein expression was detected by Western blot. Results Dioscin inhibited the activity of SW579 cells and inhibited its proliferation;it could promote the apoptosis of SW579 cells, increased the ratio of Bax/Bcl-2, and increased the expression of Caspase-3 mRNA;and inhibited cell invasion;dioscin could also down-regulate Notch 1, Jagged 1, and Hes 1 protein expression(P<0.05);and diosgenin partially reversed the effect of Jagged 1. Conclusion Dioscin has a certain inhibitory effect on thyroid carcinoma cells SW579, and its mechanism of action may be related to the inhibition of Notch 1 signaling pathway to a certain extent.
