摘 要:目的探究肝癌细胞自噬环节中Polo样蛋白激酶4(PLK4)的调控作用及相关机制。方法利用TCGA数据库探索PLK4在肝癌组织与癌旁组织中表达。Real-time PCR检测PLK4在肝癌细胞及正常肝细胞中的表达差异情况。采用小干扰RNA(siRNA)下调肝癌细胞中PLK4的表达,Western blot检测自噬相关蛋白LC3及沉默信息调节因子1(SIRT1)表达情况,免疫荧光评估LC3荧光斑点的表达水平。结果 PLK4在肝癌组织和细胞中表达升高。当PLK4下调时,自噬水平降低,SIRT1表达水平降低。结论 PLK4通过SIRT1参与了肝癌细胞自噬调控,有望成为肝癌靶向治疗中的新型分子靶点。Objective Explore the effect of PLK4 on autophagy of hepatocellular carcinoma(HCC) cells and its related mechanisms. Methods The expression of PLK4 in HCC and adjacent tissues was investigated using TCGA database. Real-time PCR was used to detect the expression differences of PLK4 in HCC cells and normal liver cells. Small interfering RNA(siRNA) was used to down-regulate the expression of PLK4 in HCC cells, the expression of autophagyrelated proteins LC3 and SIRT1 was detected by Western blot, and the level of LC3 fluorescent spots was evaluated by immunofluorescence. Results The expression of PLK4 was increased in hepatocellular carcinoma. When PLK4 was downregulated, the activity of autophagy decreased. Conclusion PLK4 is involved in autophagy regulation of liver cancer cells through SIRT1, which is expected to become a new molecular target in the therapy of liver cancer.
