摘 要:研究了槐米黄酮的纯化条件和体外抗氧化活性。结果表明,槐米黄酮经聚酰胺柱层析的最适纯化条件:黄酮上样液pH值6~7,质量浓度5 mg/mL,上样量1.5 BV,吸附流速1.0 mL/min,用3 BV去离子水清洗聚酰胺柱,4 BV解吸液80%乙醇溶液,解吸流速1.5 mL/min,聚酰胺树脂反复使用5次后进行再生处理。以Vc为阳性对照,槐米黄酮具有一定的体外抗氧化能力,其中在高浓度区间对·DPPH有较好的清除效果,且与Vc接近,对·O^(-)_(2)和·OH都有一定的自由基清除能力,但效果稍差于Vc,对Fe^(3+)有较好还原力,且在较低和较高质量浓度范围内好于Vc。The purification conditions and in vitro antioxidant activity of total flavonoids from Sophora Japonica were studied.The results showed that the optimum purification conditions of flavonoids of Sophora Japonica were as follows:The pH of sample solution was 6~7,concentration was 5 mg/mL,sample volume was 1.5 BV,the adsorption velocity was 1.0 mL/min,leaching water volume was 3,4 BV of 80%ethanol solution with 1.5 mL/min of desorption velocity best and polyamide resin regeneration treatment after repeated use of five times.Vc as the positive control,flavonoids from Sophora Japonica had certain antioxidant activity in vitro,which showed a better scavenging effect on·DPPH in the high concentration range,which was close to Vc,a certain scavenging ability on·O^(-)_(2) and·OH,which was slightly worse than Vc,and a better reducing power on Fe^(3+),which was better than Vc in the lower and higher concentration range.
