摘　　要：目的:探讨叉头框转录因子O亚族3a(FOXO3a)对人结肠癌SW620细胞的影响,并分析其可能机制。方法:将转染FOXO3a过表达的载体作用于结肠癌SW620细胞。采用MTT法检测48 h后各组细胞生长抑制率。采用免疫细胞化学检测转各组FOXO3a、CD133、八聚体结合转录因子4(OCT4)蛋白表达阳性率。采用反转录聚合酶链反应(RT-PCR)检测各组FOXO3a、CD133、OCT4 mRNA相对表达水平。采用Western blot检测各组FOXO3a、CD133、OCT4蛋白相对表达水平。采用细胞迁移侵袭实验测定各组细胞穿膜数。对各组上述指标进行比较。结果:转染FOXO3a过表达载体后,结肠癌SW620细胞生长抑制率提高,FOXO3a蛋白表达阳性率增高,CD133、OCT4 mRNA及蛋白相对表达水平降低(P<0.05),迁移侵袭能力受到抑制(均P<0.05)。结论:FOXO3a可以抑制SW620细胞的增殖,减弱肿瘤细胞迁移侵袭能力,其机制可能与降低CD133、OCT4表达有关。Objective:To investigate the effect of FOXO3a on human colon cancer SW620 cells,and analyze its possible mechanism.Methods:FOXO3a overexpression vector was transfected into SW620 cells for 48 hours.The MTT method was used to detect the cell growth inhibition rate.Immunocytochemistry was used to detect the positive rate of FOXO3a,CD133,OCT4 protein expression.RT-PCR was used to detect the relative expression levels of FOXO3a,CD133 and OCT4 mRNA.Western blot was used to detect the relative expression levels of FOXO3a,CD133 and OCT4 proteins.The cell migration and invasion experiment was used to determine the number of cells passing through the membrane.The above indicators were compared between groups.Results:After the FOXO3a overexpression vector was transfected,the growth inhibition rate of SW620 cells increased,the positive rate of FOXO3a protein expression increased,the relative expression levels of CD133,OCT4 mRNAs and proteins decreased,and the migration and invasion ability was inhibited(all P<0.05).Conclusion:FOXO3a can inhibit the proliferation of SW620 cells and reduce the invasion and migration ability of tumor cells.The mechanism may be related to the reduction of CD133 and OCT4 expression.
