摘　　要：目的:检测并比较核糖体蛋白L22(ribosomal protein L22,RPL22)在乳腺癌组织及癌旁组织中的表达,分析其在乳腺癌组织中的表达意义及其对乳腺癌预后的影响。方法:选择行乳腺癌改良根治术的62例患者,采用免疫荧光法检测乳腺癌及癌旁组织RPL22的表达,运用qRT-PCR检测乳腺癌及癌旁组织中RPL22 mRNA的表达,分析RPL22表达与临床病理参数的相关性。检索TCGA数据库,探究RPL22与患者预后的关系。结果:免疫荧光结果显示RPL22在乳腺癌组织中的表达低于癌旁组织;qRT-PCR结果显示乳腺癌组织中RPL22 mRNA的表达水平低于癌旁组织,差异有统计学意义(P<0.001)。乳腺癌组织中RPL22表达与肿瘤大小(r=-0.403)、HER-2阳性(r=-0.390、Ki67表达(r=-0.370)、淋巴结转移(r=-0.422)以及三阴性乳腺癌(TNBC)发生(r=-0.311)呈负相关。检索TCGA数据库,发现RPL22在乳腺癌组织中的表达水平明显降低,RPL22低表达组总生存率低于高表达组,差异有统计学意义(P=0.036)。结论:乳腺癌组织中RPL22表达水平较癌旁组织明显降低,并与乳腺癌不良预后有关,提示RPL22可作为乳腺癌诊断以及预后的生物学指标。Objective:To detect the content of ribosomal protein L22(PRL22)in breast cancer tissue and adjacent tissues,compare their difference,analyze its expression level in breast cancer tissue,and analyze its impact on the prognosis of breast cancer combined with clinical,so as to explore its clinical significance.Methods:The expression of RPL22 in breast cancer and adjacent tissues was observed by immunofluorescence,and the expression of RPL22 in 62 cases of breast cancer and adjacent tissues was detected by qRT-PCR.To collect the clinical and postoperative pathological data,and explore the correlation between the expression of RPL22 and clinical.By searching TCGA database,the survival rate of breast cancer patients was analyzed,and the relationship between RPL22 and prognosis was explored.Results:The results of immunofluorescence showed that the expression level of RPL22 in breast cancer tissues was significantly lower than that in adjacent tissues(P<0.001).By analyzing the clinical parameters of 62 patients with breast cancer,it was found that the low expression of RPL22 in breast cancer tissue was associated with tumor size(r=-0.403),HER-2(r=-0.390),Ki67(r=-0.370),lymph node metastasis(r=-0.422)and molecular classification of breast cancer(r=-0.311).Kaplan Meier survival curve suggested that the low expression of RPL22 was associated with the decrease of overall survival rate of BC patients(P=0.036<0.05).Conclusion:The low expression of RPL22 in breast cancer is associated with poor prognosis,which suggests that RPL22 may be a potential biomarker for the diagnosis and prognosis of breast cancer.
