摘 要:目的探讨POT1-AS1在胃腺癌(GAC)组织中的表达及其对细胞增殖的影响。方法收集2016年6月至2018年12月在西安交通大学第一附属医院普通外科手术切除的80对GAC组织及其癌旁正常组织标本(所有患者术前未接受过放、化疗等辅助治疗)、正常人胃黏膜上皮细胞(GES-1)和GAC细胞株(BGC-823、MGC-803、MKN45、SGC-7901)。RT-PCR法检测并比较POT1-AS1在GAC组织与癌旁正常组织、正常人胃黏膜上皮细胞与GAC细胞系、sh-POT1-AS1质粒转染与未转染SGC-7901细胞中的mRNA相对表达量;CCK-8实验和平板克隆实验检测POT1-AS1敲低(即sh-POT1-AS1质粒转染)对SGC-7901细胞增殖能力的影响;Western blot法检测POT1-AS1敲低对SGC-7901细胞中细胞周期相关蛋白相对表达量的影响。结果POT1-AS1在GAC组织及细胞系中均呈高表达(均P<0.05)。sh-POT1-AS1质粒转染后,SGC-7901细胞中POT1-AS1 mRNA相对表达量明显低于未转染组(P<0.05)。POT1-AS1敲低后,SGC-7901细胞增殖能力明显减弱,SGC-7901细胞克隆数亦明显减少,SGC-7901细胞中细胞周期蛋白D1(Cyclin D1)、细胞周期蛋白依赖性激酶4(CDK4)蛋白相对表达量均明显降低,而细胞周期蛋白依赖性激酶抑制因子1A(CDKI1A)蛋白相对表达量明显升高,差异均有统计学意义(均P<0.05)。结论GAC组织及细胞系中POT1-AS1 mRNA相对表达量均明显升高;在GAC细胞中敲低POT1-AS1可抑制CDK4、Cyclin D1蛋白表达并促进CDKI1A蛋白表达,进而抑制细胞增殖。Objective To investigate the expression of POT1-AS1 in gastric adenocarcinoma(GAC),and its effect on the proliferation of GAC cells.Methods Eighty pairs of GAC tissues and the corresponding paracancerous normal tissue specimens were collected from patients who did not received preoperative adjuvant treatment.The normal human gastric mucosal epithelial cells(GES-1)and GAC cell lines(BGC-823,MGC-803,MKN45,SGC-7901)were used for in vitro study.Gastric adenocarcinoma SGC-7901 cells were transfected with sh-POT1-AS1 plasmid to knockdown the POT1-AS1 expression.Real-time quantitative PCR was used to detect the expression of POT1-AS1,CCK-8 kit and plate cloning test were used to detect the proliferation ability of SGC-7901 cells,Western blotting assay was used to detect the expression levels of cell cycle-related protein in transfected and untransfected SGC-7901 cells.Results POT1-AS1 was significantly highly expressed in gastric adenocarcinoma tissues and gastric cancer cell lines(P<0.05).The relative expression of POT1-AS1 mRNA in sh-POT1-AS1-transfected SGC-7901 cells was significantly lower than that in the non-transfected SGC-7901 cells(P<0.05).Compared with untransfected SGC-7901 cells,the proliferation ability and the number of cell clones in POT1-AS1-knocked down SGC-7901 cells were significantly reduced;the expression of cell cycle protein-dependent kinase 4(CDK4)and CyclinD1 was significantly down-regulated and the expression of cyclin-dependent kinase inhibitor 1A(CDKI1A)protein was up-regulated(all P<0.05).Conclusion The expression of POT1-AS1 in GAC tissues and gastric cancer cell lines is significantly increased.Knockdown of POT1-AS1 can down-regulate CDK4,Cyclin D1 and up-regulate CDKI1A protein expression in GAC cells to inhibit GAC cells proliferation.
