摘 要:目的探讨跨膜丝氨酸蛋白酶2(TMPRSS2)在肺腺癌和肺鳞状细胞癌中的表达及其与预后的关系。方法使用Oncomine、GEPIA和UALCAN数据库分析TMPRSS2在肺腺癌和肺鳞状细胞癌中的表达水平和启动子的甲基化水平,并使用Kaplan-Meier plotter数据库分析TMPRSS2表达水平对该类患者预后的影响。基于TIMER数据库分析TMPRSS2表达与免疫细胞浸润的关系及免疫浸润对预后的影响。结果在肺腺癌和肺鳞状细胞癌中,相比正常肺组织TMPRSS2的表达量均显著降低(P<0.05),其启动子甲基化水平均出现显著增高(P<0.05)。肺腺癌中TMPRSS2的高表达有较好的预后(HR=0.49,95%CI 0.39~0.63;P=3.9×10-9),而在肺鳞状细胞癌中无相关性(HR=1.15,95%CI 0.89~1.48;P=0.28)。肺腺癌和肺鳞状细胞癌中TMPRSS2表达与多种免疫细胞浸润有关(P<0.05),B细胞和CD4+T细胞浸润水平与肺腺癌患者的预后相关(P<0.05)。结论TMPRSS2在肺腺癌和肺鳞状细胞癌组织中表达均显著下降,TMPRSS2的表达水平可能通过免疫细胞浸润影响肺腺癌患者的预后。Objective To investigate the expression of transmembrane protease serine 2(TMPRSS2)in lung adenocarcinoma(LUAD)and lung squamous cell carcinoma(LUSC)and its relationship to prognosis.Methods The Oncomine,GEPIA and UALCAN databases were applied to analyze the expression and promoter methylation levels of TMPRSS2 in LUAD and LUSC.The Kaplan-Meier plotter database was used to analyze the influence of TMPRSS2 expression level on the prognosis of patients.The TIMER database was used to analyze the relationship between TMPRSS2 expression and immune infiltration,as well as the influence of immune infiltration on the prognosis.Results Compared with the normal lung tissue,the expression of TMPRSS2 decreased but the methylation level of its promoter increased in LUAD and LUSC(P<0.05).The high expression of TMPRSS2 was associated with a better prognosis in LUAD(HR=0.49,95%CI 0.39-0.63;P=3.9×10-9),but not in LUSC(HR=1.15,95%CI 0.89-1.48;P=0.28).The expression of TMPRSS2 in LUAD and LUSC was associated with multiple immune cell infiltration(P<0.05),and the infiltration levels of B cells and CD4+T cells were significantly correlated with the prognosis of patients with LUAD(P<0.05).Conclusion The expression of TMPRSS2 decreases in both LUAD and LUSC.The expression level of TMPRSS2 may affect the prognosis of patients with LUAD through immune cell infiltration.
