参连复脉颗粒对自发性高血压大鼠心房纤维化及心房易损性的影响

摘　　要：目的探讨参连复脉颗粒对自发性高血压大鼠心房纤维化及心房易损性的影响及可能作用机制。方法将50只24周龄的雄性自发性高血压大鼠随机分为模型组、氯沙坦钾组、参连复脉低剂量组、参连复脉中剂量组和参连复脉高剂量组各10只,选择24周龄雄性正常血压京都威斯特大鼠10只为正常组。参连复脉低、中、高剂量组分别给予参连复脉颗粒0.81 g/(kg·d)、1.62 g/(kg·d)、3.24 g/(kg·d)灌胃,氯沙坦钾组给予氯沙坦钾片4.5 mg/(kg·d)灌胃,模型组和正常组给予等剂量0.9%氯化钠溶液灌胃,均连续4周。干预结束后,测量各组大鼠血压、心率,超声心动图检测左心房内径,记录各组大鼠房颤诱发率及持续时间,Masson染色观察各组大鼠左心房间质胶原纤维分布情况并计算心房间质胶原纤维容积分数,Western blot法检测各组大鼠左心房肌组织中转化生长因子β1(TGF-β1)、Smad3蛋白表达情况。结果与正常组比较,模型组大鼠SBP、DBP及房颤诱发率、心房胶原纤维容积分数、左心房组织中TGF-β1和Smad3蛋白表达量均明显增高(P均<0.05),左心房内径明显增大(P<0.05)。与模型组比较,氯沙坦钾组大鼠SBP,参连复脉中剂量组和氯沙坦钾组大鼠房颤诱发率,参连复脉各组和和氯沙坦钾组大鼠心房胶原纤维容积分数、左心房组织中TGF-β1和Smad3蛋白表达量均明显降低(P均<0.05),参连复脉中、高剂量组和氯沙坦钾组大鼠左心房内径均明显缩短(P均<0.05)。结论自发性高血压大鼠心房纤维化明显,心房易损性增加,参连复脉颗粒可能通过抑制心房组织TGF-β1/Smad3蛋白的表达,改善自发性高血压大鼠心房纤维化,预防房颤发生。Objective It is to investigate the possible mechanism of Shenlian Fumai Granule on atrial fibrosis and atrial vulnerability in spontaneously hypertensive rats and its possible action mechanism.Methods Fifty 24-week-old male spontaneously hypertensive rats were randomly divided into 5 groups:model group,losartan potassium group,Shenlian Fumai Granule low-dose group,middle-dose group,and high-dose group,each group had 10 rats.1024-week-old male wild-type WKY rats were served as the normal control group.Shenlian Fumai low,medium and high dose groups were respectively given Shenlian Fumai Granules 0.81 g/(kg·d),1.62 g/(kg·d),3.24 g/(kg·d)by gavage,the losartan potassium group was given losartan potassium tablets 4.5 mg/(kg·d)by gavage,and the model group and the normal group were given the same dose of 0.9%sodium chloride solution by gavage,all treated for 4 consecutive weeks.After the intervention,the blood pressure and heart rate of the rats in each group were measured,the inner diameter of the left atrium was measured by echocardiography,and the induction rate and duration of atrial fibrillation in each group were recorded.Masson staining was used to observe the distribution of collagen fibers in the left ventricle of each group and the volume fraction of atrial collagen fibers was calculated.Western blot was used to detect the expression of transforming growth factorβ1(TGF-β1)and Smad3 protein in the left atrial muscle tissue of rats in each group.Results Compared with the normal group,the SBP,DBP,atrial fibrillation induction rate,atrial collagen fiber volume fraction,and the expression of TGF-β1 and Smad3 protein in left atrial tissue in the model group were significantly increased(all P<0.05),and the left atrial diameter was significantly decreased(all P<0.05).Compared with the model group,the SBP of rats in the losartan potassium group,atrial fibrillation induction rate of rats in the losartan potassium group,the Shenlian Fumai medium dose group,the volume fraction of atria collagen fibers,TGF-β1 and
