摘 要:目的观察麦粒灸对薄型子宫内膜大鼠血管生成的影响,探讨其可能的作用机制。方法30只SPF级成年雌性SD大鼠随机分为对照组、模型组、麦粒灸组,每组10只。除对照组外,其余2组大鼠采用95%乙醇宫腔注射法于动情期制备薄型子宫内膜模型。麦粒灸组造模后次日麦粒灸“关元”“子宫”,每穴7壮,1次/d,对照组和模型组大鼠每日麻醉后固定,连续3个动情周期(约15 d)。干预结束后,HE染色观察大鼠子宫内膜形态,免疫组化检测大鼠子宫内膜组织血管内皮生长因子(VEGF)和血管生成素-2(Ang-2)表达,Western blot检测大鼠子宫内膜组织DLL4和Notch1蛋白表达,RT-qPCR检测大鼠子宫内膜组织VEGF、Ang-2、DLL4和Notch1 mRNA表达。结果与对照组比较,模型组大鼠子宫内膜明显变薄,腺体、血管数目明显减少,子宫内膜组织VEGF、Ang-2、DLL4和Notch1蛋白及mRNA表达明显降低,差异均有统计学意义(P<0.01);与模型组比较,麦粒灸组大鼠子宫内膜厚度明显增加,腺体、血管数目明显增多,子宫内膜组织VEGF、Ang-2、DLL4和Notch1蛋白及mRNA水平明显升高,差异均有统计学意义(P<0.05,P<0.01)。结论麦粒灸可改善子宫内膜厚度,促进子宫内膜血管生成,达到修复作用,其机制与促进薄型子宫内膜大鼠子宫内膜组织VEGF、Ang-2表达,上调Notch信号通路相关分子DLL4、Notch1水平有关。Objective To observe the effect of wheat-grain moxibustion on angiogenesis in rats with thin endometrium;To explore its possible mechanism.Methods Totally thirty healthy SPF-grade adult female SD rats were randomly divided into control group,model group,and wheat-grain moxibustion group,with 10 rats in each group.Except for the control group,rats in the other two groups were injected with 95%ethanol into the uterus to establish a thin endometrial model during estrus.The control group and the model group were fixed daily after anesthesia,and the wheat-grain moxibustion group was given wheat-grain moxibustion at the acupoints of“Guanyuan”(CV 4)and“Zigong”(EX-CA1)the next day after the model was established,7 moxa cones for each acupoint,once a day,for 3 consecutive estrus cycles(approximately 15 days).After the intervention,HE staining was used to observe the morphological changes of the rat’s endometrium;the expressions of vascular endothelial growth factor(VEGF)and angiopoietin 2(Ang-2)were detected by immunohistochemistry;the protein and mRNA expressions of Notch receptor(Notch1)and Notch ligand(DLL4)were detected by Western blot;RT-qPCR was used to detect the expressions of VEGF,Ang-2,DLL4 and Notch1 mRNA in rat endometrial tissue.Results Compared with the control group,the endometrial thickness of rats in the model group was significantly thinner;the numbers of endometrial glands and blood vessels were significantly reduced;the expressions of VEGF,Ang-2,DLL4 and Notch1 protein and mRNA in endometrial tissue were significantly reduced,with statistical significance(P<0.01).Compared with the model group,the thickness of endometrium in the wheat-grain moxibustion group was thicker;the numbers of endometrial glands and blood vessels increased significantly;VEGF,Ang-2,DLL4 and Notch1 protein and mRNA levels in endometrial tissue significantly increased,with statistical significance(P<0.05,P<0.01).Conclusion Wheat-grain moxibustion can improve endometrial thickness,promote endometrial angiogenesis,and achie
