摘 要:以二甲基亚砜、丙三醇、甲醇、蔗糖、聚乙二醇为灵芝菌种QL 02低温保藏的保护剂材料,对影响菌丝活性的关键保护剂种类进行筛选,通过响应面法对灵芝菌种QL 02号的低温保藏保护剂配方进行优化。结果表明,保护剂中甲醇含量、丙三醇含量和蔗糖含量为影响-20℃低温保藏灵芝菌株QL 02活性的关键因素;优化后的最优保护剂配方为蔗糖5.8%、甲醇8%、丙三醇4.7%。在此最优条件下,菌丝平均生长速度为5.13 mm·d^(-1)。In this study,dimethyl sulfoxide,glycerol,methanol,sucrose and polyethylene glycol were used as the protectant materials for low temperature preservation of Ganoderma lucidum strain QL 02.The key types of protectant affecting the activity of mycelia were screened,and then the formulation of the protectant for low temperature preservation of Ganoderma lucidum strain QL 02 was optimized by response surface methodology.The results showed that the contents of methanol,glycerol and sucrose in the protectant were the key factors affecting the activity of QL 02 preserved at low temperature.The optimal formula of protective agent after optimization was 5.8%sucrose,8%methanol and 4.7%glycerol.With the optimal conditions,the average growth rate of mycelia was 5.13 mm·d^(-1).
