苏格木勒-7对化疗致卵巢功能低下大鼠的卵巢功能保护机制

摘　　要：目的探究苏格木勒-7对卵巢功能低下模型大鼠的卵巢功能保护机制。方法采用48只正常性周期大鼠随机分为4组:模型组即环磷酰胺组(CTX)、模型加苏格木勒-7[0.5 g/(kg·d)]组即苏-7-0.5组、模型加苏格木勒-7[1.0 g/(kg·d)]组即苏-7-1.0组、正常组,除了正常组其他3组均连续7 d腹腔注射CTX 30 mg/(kg·d)。正常组和模型组等量生理盐水灌胃,其余给药组在造模第1天开始(建模同时)分别灌胃给予苏-7-1.0 g/(kg·d)和苏-7-0.5 g/(kg·d),每天给药1次,连续给药24 d,采用免疫组织化学法检测卵巢组织氧化应激相关蛋白8-羟基脱氧乌苷(OHdG)、4-羟基壬烯醛(HNE)、硝基酪氨酸(NTY)及卵泡发育相关细胞因子骨形成态蛋白质(BMP)-15、生长分化因子(GDF)-9的定位及表达量;用Western印迹法检测卵巢组织凋亡相关蛋白及抗氧化相关蛋白Bax、Bcl-2、超氧化物歧化酶(SOD)、血小板反应蛋白(TSP)-1、组织蛋白酶(Cathepin)D、骨髓基质细胞抗原(BST)1、补体因子-H(CFH)等的表达量。结果苏格木勒-7对大鼠血清肿瘤坏死因子(TNF)-α、白细胞介素(IL)-6、转化生长因子(TGF)-β1明显改善;Western印迹检测结果,对Bax、Bcl-2、SOD及TSP-1、CFH等显著改善。结论苏-7通过多方面、多途径来对化疗性卵巢功能低下大鼠内分泌和生殖功能起到保护作用。Objective To explore the protective mechanism of sugimule-7 on ovarian function in rats with ovarian dysfunction.Methods 48 rats with normal sexual cycle were randomly divided into model group,cyclophosphamide group(CTX),model+sugimule-7[0.5 g/(kg•d)]group(sugimule-7-0.5 group),model+sugimule-7[1.0 g/(kg•d)]group(sugimule-7-1.0 group)and normal group(NC).Except the normal group,the other three groups were injected intraperitoneally with CTX 30 mg/(kg•d)for 7 days.The normal group and the model group were given the same amount of normal saline by gavage,and the other groups were given sugimule-7-1.0 g/(kg•d)and sugimule-7-0.5 g/(kg•d)by gavage from the first day of modeling(at the same time),once a day for 24 consecutive days.The oxidative stress related proteins 8-OHdG,4-HNE,NTY and cytokines related to follicular development BMP-15 were detected by immunohistochemical method localization and expression of GDF-9.The expressions of apoptosis related proteins and antioxidant related proteins Bax,Bcl-2,SOD,TSP-1,cathepind,bst1 and CFH were detected by Western blot.Results Sugimule-7 had no effect on serum TNF-α,IL-6,TGF-β1.Western blot showed that Bax,Bcl-2,SOD,TSP-1 and CFH were significantly improved.Conclusions Sugimule-7 could protect the endocrine and reproductive function of chemotherapy-induced ovarian dysfunction rats in many ways.
