沉默lncRNA TPT1-AS1对非小细胞肺癌生物学行为的影响及机制研究

摘　　要：目的研究长链非编码RNA(lncRNA)TPT1-AS1在非小细胞肺癌组织中的表达及对沉默lncRNA TPT1-AS1非小细胞肺癌生物学行为的影响及机制。方法检测非小细胞肺癌组织和癌旁组织lncRNA TPT1-AS1表达情况,做细胞培养及lncRNA TPT1-AS1转染,分为空白组、过表达组、沉默组,过表达组转染si-TPT1-AS1-NC质粒,沉默组转染si-TPT1-AS1质粒,空白组细胞不做任何处理,转染24h后鉴定转染效率。检测各组细胞增殖、凋亡、迁移、侵袭及PTEN-PI3K-AKT信号通路蛋白表达情况。结果与癌旁组织相比,癌组织TPT1-AS1 mRNA表达量升高(P<0.05)。与空白组相比,过表达组TPT1-AS1 mRNA表达量升高,沉默组TPT1-AS1 mRNA表达量降低(P<0.05),说明转染成功。与过表达组相比,沉默组各时间点细胞增殖率、细胞迁移数、侵袭数、p-PI3K、p-AKT、MMP-2、MMP-9、Bcl-2蛋白表达量降低,凋亡率、PTEN、Bax蛋白表达量升高(P<0.05)。结论lncRNA TPT1-AS1在非小细胞肺癌组织高表达,沉默lncRNA TPT1-AS1可抑制肺癌细胞增殖、迁移、侵袭,促进凋亡,其机制可能与调控PTEN-PI3K-AKT信号通路相关。Objective To study the expression of long chain non-coding RNA(lnc RNA)TPT1-AS1 in non-small cell lung cancer and its effect of silencing lncRNA TPT1-AS1 on the biological behavior of silent non-small cell lung cancer.Methods The expression of lncRNA TPT1-AS1 in non-small and fine lung cancer tissues and adjacent tissues were detected.The cells were cultured and transfected with lncrna TPT1-AS1.The cells were divided into the blank group,the over-expression group and the silent group.The over expression group was transfected with si TPT1-AS1-NC plasmid,and the silence group was transfected with si TPT1-AS1 plasmid.The cells in the blank group were not treated any way,and the transfection efficiency was determined 24 hours after transfection.The proliferation,apoptosis,migration,invasion and expression of PTEN-PI3K-AKT signaling pathway protein were detected.Results Compared with the adjacent tissues,the expression of TPT1-AS1 mRNA in cancer tissues was increased(P<0.05).Compared with the blank group,the expression of TPT1-AS1 mRNA in the over-expression group was higher than that in the silence group(P<0.05),indicating that the transfection was successful.Compared with the over-expression group,the cell proliferation rate,cell migration number,invasion number,p-PI3K,p-Akt,MMP-2,MMP-9,Bcl-2 protein expression levels in the silence group were decreased,and the apoptosis rate,PTEN and Bax protein expression were increased(P<0.05).Conclusion lncRNA TPT1-AS1 is highly expressed in non-small cell lung cancer tissues.Silencing lncRNA TPT1-AS1 can inhibit the proliferation,migration,invasion and promote apoptosis of lung cancer cells,and the mechanism may be related to the regulation of PTEN-PI3K-AKT signaling pathway.
