摘  要:目的:研究义眼座-细胞复合体在体外的生物活性。方法:将12个大小约3 mm×3 mm×2 mm的羟基磷灰石义眼座块,随机分为两组,每组6个:实验组接种50 μl浓度为1×10 6/ml的血管内皮细胞悬液;对照组接种等量无细胞培养液;两组均放入10%完全培养基(10%胎牛血清与90%高糖培养液配制)后5、10及15 d,对两组培养基中血管内皮生长因子、前列环素及内皮素进行定量检测;20 d后做义眼座病理切片,观察义眼座内细胞生长情况。 结果:光学显微镜下观察皿内细胞正常增生,实验组三种因子在不同时间的分泌量存在差异( P<0.05);细胞因子量随时间增加而增加,第15天VEGF、PGI2、EDN1因子分泌量分别为(6 147.31±691.50)pg/ml、(104.92±6.32)pg/ml和(6.32±0.49)pg/ml,对照组未见细胞因子,实验组义眼座块内可见血管内皮细胞长入,部分细胞形成管腔样结构。 结论:体外培养的血管内皮细胞在义眼座内生长情况良好,细胞-义眼座复合体具有良好的生物活性。Objective To study the bioactivity of artificial globe pedestal-cell complex in vitro.Methods Twelve hydroxyapatite fragments of artificial globe pedestal with a size of 3 mm×3 mm×2 mm were randomly divided into two groups.The experiment group was inoculated with 50μl of suspension with a concentration of 1×10^(6) human unbilical vein endothelial cells per ml,whereas the control group was inoculated with equal amount of acellular medium.The pedestals of the two groups were cultured in 10%complete culture medium(preparation of 10%fetal bovine serum and 90%high glucose medium)for 5,10 and 15 days.The vascular endothelial growth factor,VEGF,epoprostenol,PGI2 and endothelin 1,EDN1 were measured.Furthermore,pathological section of the artificial globe pedestal in the experiment group after 20 days’culture was used to observe the cell growth.Results The microscope observation revealed that cell proliferated normally in the dish.The difference in secretion of the three factors in the experiment group was statistically significant at different time points(P<0.05).The amount of cytokines increased with time.On the 15th day after culture,the secretion of VEGF,PGI2 and EDN1 were(6147.31±691.50)pg/ml,(104.92±6.32)pg/ml and(6.32±0.49)pg/ml,respectively.No cytokine was found in the control group.Vascular endothelial cells grew into the artificial globe pedestal in the experiment group,and formed lumen-like structures.Conclusion The vascular endothelial cells grow well and proliferate significantly in the orbital fragments,which indicates that the cell-scaffold complex possesses good bioactivity.
