摘 要:为研究黄淮麦区(南片)小麦籽粒中过氧化物酶(peroxidase,POD)的活性及其等位基因在此麦区新育成小麦品种(系)中的分布情况,利用分光光度计对黄淮麦区(南片)新育成的94份小麦品种(系)的过氧化物酶活性进行测定,同时利用功能标记POD-3A1、POD-3A2、POD-7D1和POD-7D6对供试材料TaPod-A1和TaPod-D1位点的等位基因进行检测。结果表明,参试小麦材料POD活性的平均值为668.6 U·g^(-1)·min^(-1),变化范围为431.3~954.8 U·g^(-1)·min^(-1),大部分供试材料的POD活性为中等水平;在TaPod-A1位点,共检测到TaPod-A1a和TaPod-A1b两种等位基因,分布频率分别为48.9%和51.1%,携带两种等位基因材料的POD活性差异达到显著水平,TaPod-A1a与低POD活性相关,TaPod-A1b与高POD活性相关;在TaPod-D1位点,仅检测到TaPod-D1b等位基因,分布频率为100%,说明供试材料在TaPod-D1位点的多态性较为单一。本研究可为黄淮麦区(南片)小麦品质色泽改良提供参考信息。In order to study the peroxidase(POD)activity and the distribution of alleles in the newly bred wheat varieties from Huanghuai southern region,the spectrophotometer was used to determine the POD activity,and the functional markers POD-3A1,POD-3A2,POD-7D1 and POD-7D6 were used to detect allelic variation of TaPod-A1 and TaPod-D1 in 94 new wheat varieties(lines).The average POD activity of the tested wheat varieties was 668.6 U·g^(-1)·min^(-1),and the variation ranging from 431.3 to 954.8 U·g^(-1)·min^(-1).The varieties with medium POD activity account for a higher proportion.At TaPod-A1 locus,the frequency of the tested wheat varieties with TaPod-A1a allele was 48.9%,and that of TaPod-A1b allele was 51.1%.The POD activity of the TaPod-A1b is significantly higher than that of TaPod-A1a(P<0.05).TaPod-A1a is related to low POD activity,while TaPod-A1b is related to high POD activity.At TaPod-D1 locus,only TaPod-D1b alleles was detected,and the distribution frequency was 100%,indicating there was no polymorphism at TaPod-D1 locus of the test materials.This study provides reference information for the improvement of wheat flour color in quality breeding in Huanghuai southern region.
