摘　　要：目的通过芪蛭胶囊中药对脑缺血/再灌注损伤(CIRI)大鼠脑组织中mTOR与P70S6K蛋白表达影响的研究,进一步探讨芪蛭胶囊在脑损伤过程中发挥脑保护作用的分子机制。方法将大鼠随机分为假手术组(sham组)、模型组(I/R组)、雷帕霉素组、中药组;MCAO/R模型制备前进行相应抑制剂及中药干预,再灌注24h后进行NSS神经功能评分;干湿比重法评定脑组织水肿程度;TUNEL法检测缺血侧脑细胞凋亡情况;Western Blot法检测缺血侧脑组织中mTOR与P70S6K蛋白表达水平。结果与I/R组相比,中药组神经功能评分显著降低,脑水肿程度显著改善,细胞凋亡明显减少,p-mTOR、p-P70S6K表达均明显降低。结论芪蛭胶囊中药复方可减少MCAO/R模型大鼠脑水肿及细胞凋亡数,下调p-mTOR、p-P70S6K蛋白表达,强化细胞重塑能力。Objective To study the molecular mechanism of Qizhi Capsule on the expression of mTOR and P70 S6 K protein in the brain tissue of rats with cerebral ischemia/reperfusion injury(CIRI),and to further explore the molecular protective mechanism of Qizhi Capsule in the process of brain injury.Methods Rats were randomly divided into sham group,model group(I/R group),rapamycin group and traditional Chinese medicine group.Before the preparation of MCAO/R model,the corresponding inhibitors and Chinese medicine intervention were performed,and NSS neurological function score was performed after 24 h of reperfusion.The degree of brain edema was evaluated by dry-wet specific gravity method.TUNEL assay was used to detect the apoptosis of ischemic side brain cells.The expression levels of p-mTOR and p-P70 S6 K protein in ischemic brain tissues were detected by Western Blot.Results Compared with the I/R group,the neurological score was significantly reduced,the degree of brain edema was significantly improved,apoptosis was significantly reduced,and the expression of p-mTOR and p-P70 S6 K protein was significantly decreased in the TCM group.Conclusion Qizhi capsule TCM compound can reduce brain edema and cell apoptosis in MCAO/R model rats,down-regulate the expression of p-mTOR and p-P70 S6 K protein,and strengthen the ability of cell remodeling.
