摘 要:目的:观察龙葵碱对胃癌小鼠血清中肿瘤坏死因子-α(Tumor Necrosis Factor-α,TNF-α)、脂多糖受体CD14(简称CD14)、核转录因子-κB(Nuclear Factor-κB,NF-κB)和一氧化氮(Nitric Oxide,NO)的含量及瘤组织中半胱氨酸天冬氨酸蛋白酶-3(Cysteine Aspartate Proteinase-3,caspase-3)、埃兹蛋白(Ezrin Protein,Ezrin)、B淋巴细胞瘤-2基因(B-cell lymphoma-2, Bcl-2)和Bcl2-Associated X的蛋白质(Bcl2-Associated X protein, Bax)表达水平的影响,分析其抑制胃癌的作用及机制。方法:40只BALB/C小鼠于右腋下部接种胃癌细胞株悬液制作荷瘤小鼠模型,随机分成5组即模型组、龙葵碱(高、中、低)剂量组、猪苓多糖组,酶联免疫吸附剂测定(Enzyme-linked Immunosorbent Assay,ELISA)法检测各组鼠血清中TNF-α、CD14、NF-κB和NO的含量变化;实时荧光定量聚合酶链式反应(Quantitative Real-time Fluorescence Polymerase Chain Reaction,qRT-PCR)法检测Bax、Bcl-2、caspase-3、Ezrin基因表达情况。结果:高、中、低剂量的龙葵碱可增加胸腺和脾脏指数,抑制移植瘤生长;可减少鼠血清中CD14和NF-κB的含量,增加TNF-α和NO的含量水平;可增加Bax和caspase-3的基因表达,减少Bcl-2和Ezrin基因表达。在龙葵碱中剂量组中,上述变化较为明显。结论:在荷瘤小鼠模型中,龙葵碱可能通过下调CD14、NF-κB、Ezrin和Bcl-2的表达,上调TNF-α、NO、Bax和caspase-3的表达以达到抑制胃癌进展的作用。Objective: To observe the effects of solanine on TNF-α, lipopolysaccharide receptor CD14, NF-κB and NO contents in the serum of mice with gastric cancer, and its influences on the levels of caspase-3, Ezrin, Bcl-2 and Bax, then to analyze its function and mechanism of inhibitting gastric cancer. Methods: 40 BALB/C mice were inoculated with gastric cancer cell line suspension in the right axillary to make tumor-bearing mouse models, and then were randomly divided into 5 groups, namely the model group, the solanine(high,medium, and low dose) groups, and the polyporus polysaccharide group. The changes of TNF-α, CD14, NF-κB and NO levels in tumorbearing mouse serum in each group were detected by ELISA method. The expressions of Bax, Bcl-2, caspase-3, and Ezrin genes were detected by qRT-PCR method. Results: High, medium and low doses of solanine can increase the indexes of thymus and spleen, inhibit the growth of transplanted tumor, reduce the content of CD14 and NF-κB in the serum of mice, increase the content of TNF-α and NO, increase the levels of Bax and caspase-3 genes, reduce Bcl-2 and Ezrin genes expression. In the solanine middle dose group, the above changes were more obvious. Conclusion: Solanine may reduce the expression of CD14, NF-κB, Ezrin and Bcl-2, and up-regulate the expression of TNF-α, NO, Bax and caspase-3 in tumor-bearing mouse to inhibit the progression of gastric cancer.
