摘 要:目的观察宫颈癌组织中长链非编码RNA同源盒基因D-反义链1(lncRNA HOXD-AS1)、微小RNA-133a-3p(miR-133a-3p)的表达变化,并探讨其临床意义。方法选取手术切除的宫颈癌及其癌旁组织各92例份,采用qRT-PCR法检测上述组织lncRNA HOXD-AS1、miR-133a-3p,分析两指标间及其与宫颈癌临床病理参数的关系,Kaplan-Meier曲线分析不同lncRNA HOXD-AS1、miR-133a-3p表达水平宫颈癌患者术后3年总生存率,Cox回归分析法分析宫颈癌患者预后不良的影响因素。结果宫颈癌组织中lncRNA HOXD-AS1、miR-133a-3p相对表达量分别为2.064±0.644、0.947±0.305,癌旁组织分别为1.044±0.275、1.572±0.563,两者比较,P均<0.05。宫颈癌组织中lncRNA HOXD-AS1与miR-133a-3p表达呈负相关(r=-0.663,P<0.05)。lncRNA HOXD-AS1、miR-133a-3p表达与宫颈癌FIGO分期、淋巴结转移有关(P均<0.05)。lncRNA HOXD-AS1≥2.064患者(40例)和lncRNA HOXD-AS1<2.064患者(52例)术后3年总生存率分别为30.00%(12/40)、67.31%(35/52),两者比较,P<0.05;miR-133a-3p≥0.947患者(45例)和miR-133a-3p<0.947患者(47例)术后3年总生存率分别为68.89%(31/45)、34.04%(16/47),两者比较,P<0.05。多因素分析显示,FIGO分期Ⅲ~Ⅳ期、淋巴结转移、lncRNA HOXD-AS1≥2.064为宫颈癌患者预后不良独立风险因素,miR-133a-3p≥0.947为独立保护因素(P均<0.05)。结论宫颈癌组织中lncRNA HOXD-AS1表达升高,miR-133a-3p表达降低,两指标可能成为宫颈癌患者预后评估指标,lncRNA HOXD-AS1≥2.064及miR-133a-3p≥0.947时是宫颈癌患者预后不良的影响因素。Objective To observe the expression of long non-coding RNA homology cassette gene D-antisense strand 1(lncRNA HOXD-AS1) and microRNA-133a-3p(miR-133a-3p) in the cervical cancer tissues and to explore its clinical significance.Methods Ninety-two cases of cervical cancer and 92 cases of adjacent tissues were selected.LncRNA HOXD-AS1 and miR-133a-3p were detected by qRT-PCR,and the relationships between the two indexes and clinicopathological parameters of cervical cancer were analyzed.Kaplan Meier curve was used to analyze the 3-year overall survival rates of cervical cancer patients with different expression levels of lncRNA HOXD-AS1 and miR-133a-3p.Cox regression analysis was used to analyze the influencing factors of poor prognosis in patients with cervical cancer.Results The relative expression levels of lncRNA HOXD-AS1 and miR-133a-3p were 2.064±0.644 and 0.947±0.305,respectively,in the cervical cancer tissues,versus 1.044±0.275 and 1.572±0.563 in the adjacent tissues,and there was significant difference between them(all P<0.05).There was a negative correlation between lncRNA HOXD-AS1 and miR-133a-3p expression in cervical cancer(r=-0.663,P<0.05).The expression levels of lncRNA HOXD-AS1 and miR-133a-3p were related to FIGO stage and lymph node metastasis of cervical cancer(both P<0.05).The 3-year overall survival rates of patients with lncRNA HOXD-AS1≥ 2.064(40 cases) and patients with lncRNA HOXD-AS1 <2.064(52 cases) were30.00%(12/40) and 67.31%(35/52),respectively(P<0.05);the 3-year overall survival rates of patients with miR-133a-3p≥ 0.947(45 cases) and miR-133a-3p <0.947(47 cases) were 68.89%(31/45) and 34.04%(16/47),respectively(P<0.05).Multivariate analysis showed that FIGO stage Ⅲ-Ⅳ,lymph node metastasis,and lncRNA HOXD-AS1≥2.064 were independent risk factors for poor prognosis of cervical cancer patients,and miR-133a-3p≥ 0.947 was independent protective factor(all P<0.05).Conclusions The expression of lncRNA HOXD-AS1 increases,the expression of miR-133a-3p decreases in cervical cancer,a
