摘 要:目的到目前为止,肿瘤抑制基因DRR1被发现在各种组织中表达广泛,但其作用机制尚未完全揭示。本研究主要对DRR1在细胞核内与COMMD1基因之间相互的作用进行研究。方法采用免疫荧光染色的方法检测DRR1和COMMD1在HEK293T细胞核内表达情况,同时,在细胞中过表达DRR1后检测细胞核内COMMD1含量变化。结果DRR1主要在细胞核内表达,且DRR1与COMMD1在细胞核内相互结合;过表达DRR1可以促进细胞核内COMMD1的富集。结论DRR1对COMMD1在细胞核内富集具有调控作用,为DRR1的作用机制提供了新的理论依据。Objective So far,tumor suppressor gene DRR1 has been found to be widely expressed in various tissues,but its mechanism of action has not been fully revealed.In this study,the interaction between DRR1 and COMMD1 gene in nucleus was researched.Methods We determined the nuclear location of DRR1 and COMMD1 in HEK293T cells by using immunofluorescence staining.We also detected the nuclear localization of COMMD1 after DRR1 overexpression.Results DRR1 was expressed in the nucleus,and co-localization with COMMD1 in the nucleus.Nuclear COMMD1 was increased after the overexpression of DRR1 in the cells.Conclusion DRR1 promotes the nuclear enrichment of COMMD1,and it would provide a new theoretical basis for the mechanism of DRR1.
