摘  要:目的首荟通便胶囊对小鼠肠系膜微循环的影响及作用机制。方法72只昆明小鼠随机分为对照组、模型组、复方血栓组、首荟通便胶囊低剂量组、中剂量组和高剂量组,首荟通便胶囊低剂量组、中剂量组、高剂量组小鼠每日给予0.10g/kg、0.30g/kg和0.90g/kg的首荟通便胶囊灌胃,复方血栓组每日给予0.40g/kg的复方血栓灌胃,对照组和模型组给予等体积的生理盐水灌胃,连续给药7d。末次给药30min后,模型组、复方血栓组、首荟通便胶囊低剂量组、中剂量组、高剂量组分别给予0.01ml 50mg/L的去甲肾上腺素,对照组给予等剂量的生理盐水,比较各组小鼠肠系膜微动脉口径、微静脉口径、微静脉流速以及毛细血管开放数量。结果与给药前比较,给予肾上腺素刺激后,小鼠肠系膜微动脉和微静脉管径缩小,血流速度和毛细血管开放数下降(P<0.05);与对照组比较,模型组、首荟通便胶囊低剂量组、中剂量组和高剂量组小鼠肠系膜微动脉和微静脉管径缩小,血流速度和毛细血管开放数下降(P<0.05);与模型组比较,首荟通便胶囊低剂量组、中剂量组和高剂量组小鼠肠系膜微动脉和微静脉管径扩大,血流速度和毛细血管开放数增加,并呈显著的剂量依赖效应(P<0.05)。结论首荟通便胶囊可通过扩张小鼠肠系膜微血管管径、增加微血管血流速度和毛细血管开放数,改善小鼠的肠系膜微循环障碍。Objective Aim To investigate the effect of Shouhui Tongbian Capsule on Mouse Mesentery Microcirculation and Its Mechanism.Methods 72 Kunming mice were randomly divided into control group,model group,compound thrombus group,Shouhui Tongbian capsule low-dose group,medium-dose group and high-dose group.Shouhui Tongbian capsule low-dose group,middle-dose group,and high-dose group mice were given 0.10g/kg,0.30g/kg and 0.90g/kg Shouhui Tongbian capsules by gavage daily,respectively.The compound thrombus group was given 0.40g/kg compound thrombus per day by gavage,and the control group and model group were given an equal volume of normal saline gavage.After continuous administration for 7 days,the model group,compound thrombus group,Shouhui laxative capsule low-dose group,medium-dose group,and high-dose group were given 0.01ml of 50mg/L norepinephrine,and the control group was given the same dose of normal saline.The diameters of mesenteric arterioles,venules,venule flow rates and capillary openings were compared in each group.Results After adrenaline stimulation,the diameter of mesenteric arterioles and venules decreased,while the blood flow velocity and capillary opening number decreased compared with pre-administration(P<0.05).Compared with control group,the diameters of mesenteric arterioles and venules of the model group,Shouhui Tongbian capsule low-dose group,middle-dose group and high-dose group decreased,whereas the blood flow velocity and capillary opening number decreased(P<0.05).Compared with model group,the low-dose,medium-dose,and high-dose groups of Shouhui Tongbian Capsules have enlarged mesenteric arterioles and venules,increased blood flow velocity and capillary openings,and showed significant dose-dependent effects(P<0.05).Conclusion Shouhui Tongbian Capsules can improve the mesenteric microcirculation disorder of mice by expanding the diameter of the mice's mesenteric microvessels,increasing the blood flow speed of the capillaries and the number of capillary openings.
