摘 要:目的探讨萝卜硫素(SFN)对肺腺癌A549细胞侵袭迁移的抑制作用及其机制。方法将A549细胞分为干预组、模型组及对照组,干预组采用10%FBS培养基培养并给予Wnt1 5μmol/L+SFN 20μmol/L干预细胞48 h,模型组采用10%FBS培养基培养并给予Wnt1 5μmol/L干预细胞48 h,对照组采用10%FBS培养基培养48 h。划痕实验和Transwell实验检测A549细胞侵袭迁移能力。Western blotting法检测各组Wnt/β-catenin信号通路相关蛋白。结果与对照组比较,模型组细胞侵袭、迁移数量增多,与模型组比较,干预组细胞侵袭、迁移数量少(P均<0.05)。与对照组比较,模型组Wnt1、β-catenin蛋白水平升高;与模型组比较,干预组Wnt1、β-catenin蛋白水平降低(P均<0.05)。结论 SFN可抑制肺腺癌A549细胞侵袭和迁移,可能是通过抑制Wnt/β-catenin信号通路实现的。Objective To investigate the inhibitory effects of sulforaphane(SFN)on the migration and invasion of lung adenocarcinoma A549 cells.MethodsA549 cells in the logarithmic growth phase were cultured in the cell incubator with SFN at concentrations of 0,5,10,20,40 and 80 mol/L for 48 h,and cell viability was detected by MTT assay.A549 cells were divided into the control group,model group and intervention group.The cells in the intervention group were cultured in 10%FBS medium and treated with 5μmol/L Wnt1+20μmol/LSFN for 48 h,the cells in the model group were cultured in 10%FBS medium and treated with 5μmol/L Wnt1 for 48 h,and cells in the control group were cultured in 10%FBS medium for 48 h.The effects of sulforaphane on the invasion and migration of A549 cells were detected by Scratch test and Transwell test.The expression of Wnt/β-catenin signaling pathway-related proteins in each group were detected by Western blotting.ResultsCompared with 0 mol/L SFN intervention,the viability of A549 cells decreased after 5,10,20,40 and 80 mol/L SFN intervention(all P<0.05).Compared with the control group,the migration and invasion cells increased in the model group;compare with the model group,the migration and invasion cells decreased in the intervention group(all P<0.05).Compared with the control group,the levels of Wnt1 andβ-catenin increased in the model group;compared with the model group,Wnt1 andβ-catenin protein levels decreased in the intervention group(all P<0.05).ConclusionSFN could inhibit the invasion and migration of lung adenocarcinoma A549 cells by blocking the Wnt/β-catenin signal pathway transduction.
