摘  要:目的:探讨刺梨介导核因子E2相关因子2(nuclear factor erythroid 2-related factor 2,Nrf2)信号通路对过度训练大鼠骨骼肌氧化应激损伤防护的分子机制。方法:SPF级SD大鼠45只,随机分为安静对照组(C组),大强度运动组(HT组)、低剂量刺梨大强度运动组(L-HT组)、高剂量刺梨大强度运动组(H-HT组)。以跑台运动创建过度训练模型,L-HT、H-HT组分别以100 g/kg/d和200 g/kg/d的剂量灌胃刺梨原液,C组、HT组灌胃等剂量的生理盐水,6周大强度跑台运动,运动结束后次日处死,取血清和腓肠肌,分别测定各组大鼠血清超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、丙二醛(MDA)的水平、组织中Nrf2通路相关蛋白Nrf2、Kelch样环氧氯丙烷相关蛋白-1(Keap1)、血红素氧合酶1(HO-1)和凋亡蛋白B淋巴细胞瘤因子-2(Bcl-2)、Bcl-2相关X蛋白(Bax)的相对表达水平(/β-actin)。结果:1)与C组相比,HT组血清中GSH浓度降低、SOD活性下降以及MDA含量增多(P<0.01),组织中Nrf2通路相关蛋白Nrf2表达水平降低(P<0.05),Keap1没有显著性差异(P>0.05)、HO-1表达水平下降(P<0.05),Bcl-2表达水平下降(P<0.05),Bax表达升高(P<0.05),Bcl-2/Bax值下降(P<0.05)。2)与HT组相比,L-HT、H-HT组血清中GSH浓度上升、SOD活性增强以及MDA含量降低(P<0.05或P<0.01),组织中Nrf2通路相关蛋白Nrf2、HO-1表达水平升高(P<0.05或P<0.01),Keap1表达水平没有明显变化(P>0.05),Bcl-2表达水平升高(P<0.05),Bax表达下降(P<0.05),Bcl-2/Bax值增加(P<0.05)。结论:补充刺梨可介导Nrf2信号通路上调Nrf2、HO-1蛋白的表达,发挥抗氧化应激作用,进而缓解过度训练所致的大鼠骨骼肌氧化应激和过度凋亡,保护骨骼肌结构或功能的稳定。Objective:To explore the molecular mechanism of rosa roxburgh mediated nuclear factor E2-related factor 2(NRF 2)signaling pathway in protecting skeletal muscle from oxidative stress injury in overtrained rats.Methods:45 SPF SD rats were randomly divided into quiet control group(C group),high intensity exercise group(HT group),low-dose Rosa high intensive exercise group(L-HT group)and high-dose rosa high intensive exercise group(H-HT group).The overtraining model was established by treadmill exercise.The L-HT and H-HT groups were given 100g/kg/d and 200g/kg/d of rosa stock solution respectively,and the C and HT groups were given the same dose of physiological saline.After 6 weeks of intensive treadmill exercise,they were killed the next day after exercise,and serum and gastrocnemius were collected.The levels of superoxide dismutase(SOD),glutathione(GSH)and malondialdehyde(MDA)in serum,Nrf2 pathway related protein,Kelch-like epichlorohydrin related protein-1(Keap1),heme oxygenase-1(HO-1),apoptosis protein B lymphocyte tumor factor-2(Bcl-2)and Bcl-2 related X protein in tissues were measured respectively Results:1)Compared with group C,the concentration of GSH in serum,SOD activity and MDA content in HT group decreased(P<0.01),and the expression level of Nrf2 pathway related protein in tissues decreased(P<0.05),but there was no significant difference in Keap1(P>0.05),HO-1 expression level decreased(P<0.05),Bcl-2 expression level decreased(P<0.05);Bax expression level increased(P<0.05);Bcl-2/Bax expression level increased(P<0.05).2)Compared with HT group,GSH concentration in serum,SOD activity and MDA content in L-HT and H-HT groups increased(P<0.05 or P<0.01),and the expression level of Nrf2 and HO-1 in tissues increased(P<0.05 or P<0.01),but the expression level of Keap1 did not change significantly(P>0.05).Conclusion:Supplementing rosa roxburgh can mediate Nrf2 signaling pathway to up-regulate the expression of Nrf2 and HO-1 proteins and exert the effect of anti-oxidative stress,thereby alleviating oxidative stres
