摘  要:目的探讨Tnn基因对小鼠听觉功能的影响。方法①取2月龄、3月龄的C57BL/6野生型(wild-type,WT)小鼠各3只进行耳蜗冰冻切片及免疫荧光染色,观察Tnn基因在小鼠耳蜗的表达;②各取8只野生型WT小鼠及Tnn基因敲除小鼠(Tnn+/-小鼠)在1月龄、2月龄、3月龄进行短声(click)及短纯音(tome burst,tb),听性脑干反应(ABR)测试并比较结果。结果①免疫荧光显示Tnn基因在2月龄、3月龄WT小鼠耳蜗中均有表达,且主要表达于螺旋神经节神经元,与Ⅲ型β-tubulin共定位;②Tnn+/-小鼠在3月龄时,click-ABR阈值较WT型升高小鼠(分别为41.88±9.64和23.13±2.59 dB SPL,P<0.01),tb-ABR阈值4、8、16、24、32 kHz(分别为45.00±17.73、44.38±18.60、45.63±18.60、50.00±20.70、54.38±19.90 dB SPL)较WT小鼠(分别为21.25±2.31、21.87±2.59、21.25±2.31、22.50±2.67、24.38±1.77 dB SPL)均升高(P<0.05或P<0.01)。结论Tnn基因对维持小鼠听功能有重要作用,该蛋白表达异常可使小鼠听功能受损,推测其可能影响螺旋神经节神经元的正常发育,影响神经元细胞的迁移或神经突向外生长,从而引起听觉功能的异常。Objective To study the effect of Tnn on hearing function in mice by utilizing Tnn knock-out mouse.Methods①Three C57BL/6 mice were selected from different developmental stages of P2m and P3m(Ppostnatal day)each and used for cochlear frozen section and immunofluorescence staining.②Eight WT and 8 Tnn KO C57BL/6 mice were selected from different developmental stages of P1m,P2m and P3m separately.Click and tone burst ABRs were tested independently to evaluate the function of auditory nerve.Thresholds of Tnn+/-and WT mice were compared by click ABR and tb ABR.Results①Immunofluorescence showed that Tnn was expressed in the mice cochleae especially at SGNs and co-localised with ClassⅢβ-tubulin.②The hearing function of Tnn+/-mice impaired compared with WT mice,which showed the increased thresholds of click ABRs(21.13±2.59,41.88±9.64,P<0.01)at P3m and tb-ABRs at P3m of 4 kHz(21.25±2.31,45.00±17.73,P<0.01),8 kHz(21.87±2.59,44.38±18.60,P<0.05),16 kHz(21.25±2.31,45.63±18.60,P<0.01),24 kHz(22.50±2.67,50.00±20.70,P<0.01)and 32 kHz(24.38±1.77,54.38±19.90,P<0.01),respectively.Conclusion Tnn plays an essential part in the development of spiral ganglion neurons and the maintenance of hearing function.Abnormal expression of this protein may affect the neurite outgrowth,cell migration of spiral ganglion neurons or adhesion modulation function,and furthermore,may affect the function of auditory system.
