摘  要:利用pH示差法和实时荧光定量PCR测定红肉苹果夏红肉、红色之爱、克孜阿尔玛果实5个发育时期果皮中花青苷含量及其合成相关结构基因和转录因子的表达水平。结果表明:红肉苹果分别在盛花后30 d和60 d花青苷含量达到最高之后逐渐降低,在果实进一步成熟时其含量略有回升。通过实时荧光定量PCR测定红肉苹果果皮花青苷相关结构基因和转录因子的表达量,初步推测果皮花青苷积累与结构基因F3H、CHI和转录因子MYB10、bLHLH33的高水平表达有关。In this experiment,the pH difference method and real-time fluorescent quantitative PCR were used to determine the anthocyanin content in the peel of the red flesh apple,summer red flesh,"red love"and Kiz Alma fruits,and the expression level of their synthesis related structural genes and the transcription factor was also determined.The results showed that the anthocyanin content of red apples decreased gradually at 30 d and 60 d after blooming,and the content rose slightly when the fruit matured.The expression of anthocyanin related structural genes and transcription factors in red flesh apple peel was measured by real-time fluorescent quantitative PCR.It was preliminarily inferred that anthocyanin accumulation was related to the high-level expression of structural genes F3H,CHI and transcription factors MYB10,bLHLH33.
