摘  要:目的探讨miR-21、miR-221、miR-106a在儿童骨肉瘤组织中的表达及临床意义。方法收集儿童骨肉瘤患者40例作为研究组,在术中收集其病灶标本,选择同期收治的20例多指畸形患者的软骨组织作为对照组,采用免疫组化检测两组患者病灶组织中miR-21、miR-221、miR-106a蛋白表达情况。结果免疫组化分析结果表明:儿童骨肉瘤组织中miR-21、miR-221、miR-106a的相对表达[(7.74±1.89)、(5.34±1.43)、(3.88±1.06)]显著高于正常软骨组织,差异具有统计学意义(P<0.05);骨肉瘤组织miR-21、miR-221、miR-106a表达与Enneking临床分期及发生肺转移显著相关(P<0.05),但与性别、年龄、发病部位无关(P>0.05);多元Logistic回归分析结果表明,在校正性别、年龄、发病部位和肺转移等混杂性因素后,miR-21、miR-221、miR-106a水平升高仍是骨肉瘤发病的独立危险因素(P<0.05)。结论miR-21、miR-221、miR-106a可能参与了儿童骨肉瘤的疾病发展及转移过程,其机制有待进一步研究。Objective To explore the expression and clinical significance of miR-21,miR-221,miR-106 a in childhood osteosarcoma.Methods 40 children with osteosarcoma were selected as the research group.Collect specimens of their lesions during the operation.Select the cartilage tissue of 20 patients with polydactyly in the same hospital as the control group Immunohistochemistry was used to detect the expression of miR-21,miR-221 and miR-106 a in the lesion tissues of the 2 groups of patients.Results The results of immunohistochemical analysis showed that the relative expression of miR-21,miR-221 and miR-106 a in childhood osteosarcoma tissue[(7.74±1.89),(5.34±1.43),(3.88±1.06)]was significantly higher than that of normal cartilage,The difference was statistically significant(P<0.05);the expression of miR-21,miR-221,miR-106 a in osteosarcoma tissues was significantly increased in Enneking clinical stageⅡB,Ⅲand lung metastasis osteosarcoma tissues(P<0.05),but has nothing to do with gender,age,and location of onset(P>0.05);the results of multiple logistic regression analysis showed that after adjusting for confounding factors such as gender,age,location of onset,and lung metastasis,miR-21,miR-221.Increased levels of miR-106 a are still independent risk factors for the onset of osteosarcoma(P<0.05).Conclusion miR-21,miR-221,miR-106 a may be involved in the development and metastasis of childhood osteosarcoma,and the mechanism needs to be further studied.
