摘 要:为探究石莼醇提物基于细胞膜损伤的抑菌作用,检测了其对金黄色葡萄球菌、枯草芽孢杆菌、大肠杆菌、沙门氏菌的抑菌性能,测定了不同质量浓度石莼醇提物对供试菌生长、核酸和蛋白质的泄露量、培养液电导率的影响,使用流式细胞仪、扫描电镜、透射电镜观察金黄色葡萄球菌菌体损伤率和超微结构的变化.结果表明:石莼醇提物可显著抑制金黄色葡萄球菌、枯草芽孢杆菌、大肠杆菌、沙门氏菌的正常生长,尤以对金黄色葡萄球菌的抑制作用最强,其最低抑制浓度(MIC)、最低杀菌浓度(MBC)分别为1.56、3.13 mg·mL^(-1);2×MIC的石莼醇提物可增大菌体细胞膜通透性,导致菌体内核酸、蛋白质的大量泄露,培养液的电导率极显著上升(P<0.01),菌体细胞膜严重萎缩甚至裂解,菌体损伤率可达96.42%.综上,石莼醇提物通过破坏细胞膜的完整性达到抑菌效果.In order to investigate the antibacterial function of Ulva lactuca ethanol extracts by destroying cell membrane,its antibacterial activity against Staphylococcus aureus,Bacillus subtilis,Escherichia coli and Salmonella typhimurium was tested.Its impacts on the growth of bacteria,extracellular nucleic acid and protein contents,and electrical conductivity of the cultivation media were determined,and the damage rate and ultrastructural changes of S.aureus were observed by flow cytometer,scanning electron microscope and transmission electron microscope.The results showed that U.lactuca ethanol extracts exhibited excellent antimicrobial effects against the 4 bacteria tested,with the most obvious inhibitory effect being against S.aureus,whose minimal inhibitory concentration(MIC)and minimal bactericidal concentration(MBC)were 1.56 and 3.13 mg·mL^(-1),respectively.Further test indicated that 2×MIC of U.lactuca ethanol extracts was able to destroy the integrity of bacterial plasma membrane,resulting in the leakage of intracellular substance including nucleic acids and proteins of bacteria,and increases of the electrical conductivity of the culture and 96.24% of the damage rate.Overall,these results demonstrated that U.lactuca ethanol extracts could achieve antibacterial effect by destroying the integrity of cell membrane.
