摘  要:目的探讨CD47在维生素E琥珀酸酯诱导人宫颈癌Hela细胞凋亡过程中的作用。方法实验采用的细胞系是人宫颈癌Hela细胞,分为空白对照、VES处理(10μg/ml VES作用24h)、CD47抑制剂处理细胞和VES处理、CD47过表达转染细胞和VES处理、空载体转染细胞和VES处理共5组。流式细胞术检测各处理组细胞的凋亡率;Westernblot法检测各处理组细胞蛋白CD47、caspase-3和PARP活化片段的表达。结果 10μg/ml VES处理Hela细胞后,与空白对照组相比,CD47表达水平降低(P<0.05),细胞凋亡率由3.46%增加到15.5%(P<0.05),caspase-3活化片段表达和PARP蛋白剪切明显增强(P<0.05);与VES处理组相比,CD47抑制剂作用Hela细胞使凋亡率增加到21.7%(P<0.05),caspase-3活化片段的表达和PARP蛋白的剪切进一步提高(P<0.05);与空载体转染组相比,细胞转染CD47过表达质粒后,凋亡率由16.6%降低为8.04%(P<0.05),caspase-3活化片段表达和PARP蛋白的剪切有所降低(P<0.05)。结论 CD47可能参与了VES诱导的Hela细胞凋亡之后产生的结果。Objective To explore the role of CD47 in vitamin E succinate(VES)-induced apoptosis of Hela cells.Methods The cell line used in this experiment was human cervical cancer Hela cells, which were divided into five groups:blank control, VES treatment(10 μg/ml VES for 24 h), CD47 inhibitor treated cells and VES treatment, transfected cells with CD47-overexpression and VES treatment, empty vector transfected cells and VES treatment. Flow cytometry was used to detect the apoptotic rate of cells while Western blot was used to detect the expressions of CD47, caspase-3 and PARP activation fragments. Results After Hela cells were treated with 10 μg/ml VES, compared with the blank control group, the expression level of CD47 decreased(P<0.05), and the apoptotic rate increased from 3.46% to 15.5%(P<0.05). The expressions of caspase-3 activation fragments and PARP protein cleavage were significantly enhanced(P<0.05). Compared with the VES treatment group, CD47 inhibitor increased the apoptotic rate to 21.7%(P<0.05), and the expressions of caspase-3 activated fragments and PARP protein cleavage were further increased(P<0.05). Compared with the vector transfection group, the apoptotic rate was reduced from 16.6% to 8.04%(P<0.05), and the expressions of caspase-3 activation fragments and the shearing of PARP protein were reduced(P<0.05) after the cells were transfected with the CD47-overexpressed plasmid. Conclusion CD47 may be involved in Hela cell apoptosis induced by VES.
