摘 要:目的研究大豆异黄酮对环磷酰胺所致小鼠肝损伤的影响,并探讨其作用机制。方法将60只小鼠随机分为正常组、模型组、大豆异黄酮低剂量组(100 mg/kg)、大豆异黄酮中剂量组(200 mg/kg)和大豆异黄酮高剂量组(400 mg/kg)。除空白组外,其他各组按环磷酰胺50 mg/kg进行腹腔注射,共5 d,正常组给予等量生理盐水。造模D2开始,大豆异黄酮各剂量组分别灌胃给药,其余两组灌胃等体积羧甲基纤维素钠溶液,连续30d。末次给药24h后,取血和肝组织。分光光度法测定小鼠血清中的谷草转氨酶(AST)、谷丙转氨酶(ALT)的活性及肝组织中的丙二醛(MDA)、一氧化氮(NO)、谷胱甘肽(GSH)的含量;酶联免疫吸附法(ELISA)检测肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、核因子(NF-κB)表达水平;取肝左叶进行HE染色行病理学观察。结果与正常组相比,模型组小鼠血清中AST、ALT活性增强(P<0.05,P<0.01),肝组织中GSH含量降低(P<0.01),MDA、NO、TNF-α、IL-1β、NF-κB水平均升高(P<0.01);与模型组相比,大豆异黄酮中、高剂量组小鼠血清中ALT活性及肝组织NF-κB含量降低(P<0.05, P<0.01),大豆异黄酮各剂量组血清中AST及肝组织MDA、NO、TNF-α、IL-1β水平均降低(P<0.05, P<0.01),GSH含量均升高(P<0.05, P<0.01);HE染色显示肝组织形态改变明显减轻。结论大豆异黄酮对环磷酰胺所诱导的小鼠肝损伤有明显改善作用,此作用的主要机制可能与抗氧化、清除自由基和抗炎作用有关。Objective To study the effect of soybean isoflavones on liver injury induced by cyclophosphamide in mice and explore its mechanism. Methods Sixty mice were randomly divided into normal group, model group, low dose soybean isoflavones group(100 mg/kg body weight), medium dose soybean isoflavones group(200 mg/kg) and high dose soy isoflavones group(400 mg/kg). Except for mice in the normal group, mice in all other groups were intraperitoneally injected with cyclophosphamide 50 mg/kg body weight for 5 consecutive days, and the normal group was given the same amount of saline. Starting from the 2 nd day of modeling, each dose group was given soybean isoflavones orally for 30 consecutive days,and the other two groups were intragastrically given an equal volume of sodium carboxymethyl cellulose solution. Blood and liver tissues were taken from mice 24 h after the last administration. Serum activity of AST and ALT and the content of MDA,NO and glutathione(GSH) in the liver were determined by spectrophotometric methods. Levels of tumor necrosis factor(TNF-α), interleukin-1(IL-1β), and nuclear factor(NF-κB) were measured by ELISA. The left lobe of the liver was taken for HE staining for pathological observation. Results Compared with mice in the normal group, serum AST and ALT activities were enhanced(P<0.05, P<0.01), liver GSH content was decreased(P<0.01), and MDA, NO, TNF-α, IL-1β,NF-κB levels were increased in mice in the model group(P<0.01). Compared with mice in the model group, levels of ALT and NF-κB in mice in the middle and high dose soybean isoflavones groups were decreased(P<0.05, P<0.01). Levels of AST, MDA, NO, TNF-α and IL-1βin mice in the soybean isoflavones groups were all decreased(P<0.05, P<0.01)and the level of GSH in the liver were increased(P<0.05, P<0.01). HE staining showed that the morphological changes of the liver were significantly reduced. Conclusion Soybean isoflavones significantly improved liver injury induced by cyclophosphamide in mice.
