摘 要:目的采用超高效液相色谱-四极杆-静电轨道场离子阱质谱(UPLC-Q-Exactive-Orbitrap-MS)技术对新癀片中的化学成分进行鉴定分析。方法对新癀片粉末进行加热回流提取,采用Waters ACQUITY UPLC HSS T3 C18色谱柱(1.7μm,2.1 mm×150 mm),以0.1%甲酸水溶液-乙腈为流动相,梯度洗脱,流速0.3 mL·min^(-1),柱温40℃,进样量5μL;采用ESI源在正、负离子模式下分别采集数据。根据化合物精确质荷比及二级碎片离子信息,结合参考文献数据,鉴定新癀片水提液中的主要化学成分。结果从新癀片水提液中鉴定出54个化学成分,包括皂苷、胆酸、酚酸、香豆素等各类化合物。结论本研究首次采用UPLC-Q-Exactive-Orbitrap-MS技术研究新癀片中的化学成分,为新癀片成分及机制层面的研究奠定基础。Objective To determine the chemical compositions of Xinhuang tablets by UPLC-QExactive-Orbitrap-MS.Methods The powder of Xinhuang tablets was extracted via heating and refluxing.The analysis was performed on a Waters ACQUITY UPLC HSS T3 C18(1.7μm,2.1 mm×150 mm)column.The mobile phase consisted of 0.1%formic acid water solution and acetonitrile at a flow rate of 0.3 mL·min^(-1).The column temperature was 40℃and the injection volume was 5μL.The mass spectrometry was acquired with an ESI ion source in both positive and negative ion modes.The components were identified by the precise mass-to-charge ratio,secondary fragmentation and other information with literature data.Results Totally 54 compounds were identified,including saponins,bile acids,phenolic acids,and coumarins,etc.Conclusion The chemical components of Xinhuang tablets are studied by UPLC-Q-Exactive-Orbitrap-MS for the first time,providing scientific foundation for the study of the compositions and mechanism of Xinhuang tablets.
