摘 要:食管癌是全球第八大癌症,也是第六大最常见的癌症相关死亡原因。尽管近年来在手术联合放化疗方面取得了一些进展,但食管癌的预后仍然很差。食管癌发生的细胞和分子机制也尚待阐明。一些理化因素,如紫外线和γ射线,与人类癌症的发生密切相关。这些因素造成的DNA损伤是癌变的首要步骤。DNA损伤的有效修复依赖于进化中形成的DNA修复工具。受损的DNA复制如得不到有效修复,会导致基因突变,进而导致蛋白质改变。癌基因、肿瘤抑制基因或细胞周期控制基因的突变,可产生具有明显增殖优势的克隆细胞群。DNA复制和DNA损伤修复信号通路的基因高表达与食管癌患者的不良预后相关。本文总结DNA损伤与修复在食管癌中的研究进展。Esophageal cancer is the eighth most common cancer and the sixth most common cause of cancer-related death worldwide.Despite recent advances in the development of surgical techniques combining radiotherapy and chemotherapy,the prognosis of esophageal cancer remains poor.The cellular and molecular mechanisms that contribute to the pathogenesis of esophageal cancer are not yet well understood.A number of chemicals and physical agents,such as ultraviolet and gamma rays,have been implicated in the etiology of human cancer.The production of DNA damage(also known as DNA adduction or damage)caused by these agents is an important first step in the process of cancer.It has produced DNA repair tools in evolution process,which can effectively repair damaged DNA.Impairment of DNA replication may lead to gene mutations,which in turn lead to Protein-altering.oncogenes,mutations in tumor-suppressor genes or genes that control the cell cycle may give rise to clonal cell populations with significant proliferative advantages.High gene expression of DNA replication and DNA damage repair signaling pathways in esophageal cancer are associated with poor prognosis in the patients.This review summarizes the progress of DNA damage and repair in esophageal cancer.
