摘  要:目的:探讨MicroRNA-151靶向调控SYF2抑制胃癌细胞生长和迁移。方法:选取行胃癌根治术患者102例,采用qRT-PCR检测胃癌组织和癌旁组织中MicroRNA-151表达,转染SGC7901胃癌细胞系,通过CCK-8、Western blotting和克隆形成实验检测细胞生存能力、Cyclin D1、PCNA和SYF2蛋白表达和细胞集落形成能力。在SGC7901胃癌细胞系建立抑制MicroRNA-151和沉默SYF2的表达,通过细胞划痕和Transwell法实验检测细胞侵袭和迁移情况。结果:胃癌组织中MicroRNA-151表达显著高于癌旁组织的表达(P<0.05);胃癌患者组织中的MicroRNA-151表达与肿瘤的大小、TNM分期、淋巴转移转移有关(P<0.05);MicroRNA-151组的72h的细胞生存能力显著高于对照组(P<0.05);MicroRNA-151组的集落形成能力比对照组明显增加(P<0.001);MicroRNA-151组中Cyclin D1、PCNA、SYF2蛋白表达显著高于对照组;MicroRNA-151抑制组的细胞迁移能力显著低于对照组(P<0.001),抑制MicroRNA-151+sh-SYF2组的细胞迁移能力明显低于MicroRNA-151抑制组(P<0.05)。MicroRNA-151抑制组的细胞侵袭能力明显低于对照组(P<0.001),抑制MicroRNA-151+sh-SYF2组的细胞侵袭能力明显低于MicroRNA-151抑制组(P<0.05)。结论:MicroRNA-151在胃癌组织中过表达,上调SYF2水平与胃癌患者较差的临床表现和预后相关。抑制MicroRNA-151的表达,降低SYF2与Cyclin D1相互作用,抑制了胃癌的细胞活力、集落形成、侵袭和迁移能力。Objective:To investigate the microRNA-151 targeted regulation of SYF2 to inhibit the growth and migration of gastric cancer cells.Methods:A total of 102 patients undergoing radical gastrectomy were selected,and qRT-PCR was used to detect the expression of MicroRNA-151 in gastric cancer tissues and adjacent tissues.The cells were transfected with SGC7901 gastric cancer cell line,and cell survival was detected by CCK-8,Western blotting and cloning formation experiments.Capacity,Cyclin D1,PCNA and SYF2 protein expression and cell colony formation capacity.In addition,SGC7901 gastric cancer cell line was established to inhibit the expression of MicroRNA-151 and silence SYF2,and the cell invasion and migration were detected by cell scratch and Transwell experiments.Results:The expression of MicroRNA-151 in gastric cancer tissues was significantly higher than that in adjacent tissues(P<0.05);the expression of MicroRNA-151 in tissues of gastric cancer patients was significantly correlated with tumor size,TNM stage,lymphatic metastasis(P<0.05);The 72h cell viability of the MicroRNA-151 group was significantly higher than that of the control group(P<0.05);The colony forming ability of the MicroRNA-151 group was significantly increased compared with the control group(P<0.001);In the MicroRNA-151 group The protein expression of Cyclin D1,PCNA and SYF2 was significantly higher than that of the control group;the cell migration ability of the MicroRNA-151 inhibition group was significantly lower than that of the control group(P<0.001),and the cell migration ability of the MicroRNA-151+sh-SYF2 group was significantly lower MicroRNA-151 inhibition group(P<0.05).The cell invasion ability of the MicroRNA-151 inhibition group was significantly lower than that of the control group(P<0.001),and the cell invasion ability of the MicroRNA-151+sh-SYF2 group was significantly lower than that of the MicroRNA-151 inhibition group(P<0.05).Conclusion:MicroRNA-151 is overexpressed in gastric cancer tissues,and up-regulating SYF2 level is assoc
