摘  要:目的分离和鉴定外泌体并将硬脂酞辅酶A去饱和酶1(SCD-1)-小干扰RNA(siRNA)载入外泌体,探究SCD-1-siRNA外泌体对甲状腺未分化癌(ATC)Hth7和人甲状腺癌细胞(FRO)存活的影响。方法先验证ATC细胞和正常甲状腺细胞中SCD-1表达差异,再从人胚胎肾细胞(HEK293)细胞中分离外泌体并对外泌体的形态结构和ATC细胞摄取情况进行鉴定。通过电穿孔的方式将SCD-1-siRNA载入外泌体完成SCD-1-siRNA外泌体的构建,并分析SCD-1-siRNA外泌体对ATC Hth7和FRO细胞存活的影响。结果在ATC Hth7和FRO细胞中SCD-1表达明显高于甲状腺滤泡上皮Nthy细胞;成功分离外泌体并构建SCD-1-siRNA外泌体。SCD-1-siRNA外泌体可进入Hth7和FRO细胞,抑制SCD-1表达,抑制细胞存活,促进细胞死亡。结论SCD-1-siRNA外泌体可抑制ATC细胞的存活,SCD-1-siRNA外泌体可能作为ATC潜在的治疗方案。Objective To isolate and identify exosomes and load SCD-1-siRNA into exosomes,to explore the effects of SCD-1-siRNA exosomes on the survival of anaplastic thyroid carcinoma(ATC)Hth7 and FRO cells.Methods The difference in SCD-1 expression between ATC cells and normal thyroid cells was first verified,and then exosomes were isolated from HEK293 cells and their morphological structure and ATC cell uptake were identified.SCD-1-siRNA was loaded into exosomes by electroporation to complete the construction of SCD-1-siRNA exosomes.The effects of SCD-1-siRNA exosomes on the survival of ATC Hth7 and FRO cells were analyzed.Results The expression of SCD-1 in ATC Hth7 and FRO cells were significantly higher than that of thyroid follicular epithelial Nthy cells,the exosomes were successfully isolated and SCD-1-siRNA exosomes were constructed.SCD-1-siRNA exosomes can enter Hth7 and FRO cells,inhibit SCD-1 expression,inhibit cell survival,and promote cell death.Conclusion SCD-1-siRNA exosomes can inhibit the survival of ATC cells.SCD-1-siRNA exosomes may be a potential treatment option for ATC.
