摘  要:目的:分析癫痫小鼠内嗅皮层γ-氨基丁酸能神经元电生理变化及左乙拉西坦(Lev)干预对其的影响。方法:通过绿色荧光蛋白转基因(FVB-Tg)将24只癫痫小鼠分为模型组、实验组及对照组3组,每组8只,其中实验组小鼠腹腔注射东莨菪碱[200 mg/(kg·d),2次/d],模型组及对照组小鼠灌胃给药为0.9%的氯化钠(NaCl)溶液。所有受试小鼠于造模结束后直接断头取脑制作脑片,于人工脑脊液中孵育结束后,通过膜片钳技术行全细胞记录。比较3组小鼠大脑内嗅皮层γ-氨基丁酸能神经元4次动作电位的静息膜电位(Vr_(1-4))与阈电位(Vts_(1-4))的差值(Vts-Vr_(1-4))、绝对不应期(ARP_(1-4))及动作电位间距(ISI_(1-4))。结果:模型组小鼠的ARP_(1-4)明显长于对照组及实验组,差异有统计学意义(t=3.551,t=4.052,t=3.854,t=4.202;P<0.05),3组小鼠的ARP_(1-4)的比较,差异均有统计学意义(F=7.96,F=4.65,F=5.06,F=8.13;P<0.05)。实验组小鼠Vts-Vr_(1-4)明显短于模型组及对照组,差异有统计学意义(t=4.215,t=4.551,t=3.965,t=4.023;P<0.05)。模型组小鼠ISI_(1-4)明显长于对照组,差异有统计学意义(t=3.662,t=3.564,t=3.266,t=3.537;P<0.05);3组小鼠ISI_(1-4)明显长于对照组及实验组,差异有统计学意义(F=8.84,F=6.22,F=11.28,F=13.42;P<0.05)。结论:Lev干预可使癫痫小鼠内嗅皮层γ-氨基丁酸能神经元Vts-Vr、ARP、ISI明显缩短,显著改善其内嗅皮层γ-氨基丁酸能神经元的编码能力及兴奋性。Objective:To analyze the electrophysiological changes ofγ-aminobutyric neurons in endo olfactory cortex of epileptic mice and the effect of Levetiracetam(Lev)intervention on them.Methods:24 epileptic mice were divided into three groups by green fluorescent protein transgene(FVB TG):model group,experimental group and control group,with 8 mice in each group.The mice of experimental group were injected with scopolamine[200 mg/(kg•d),twice a day],and the mice of model group and control group were administered by gavage with 0.9%sodium chloride(NaCl),respectively.After modeling was completed,all mice were decapitated directly to make brain slices.After incubation was completed in artificial cerebrospinal fluid,whole cell recording was performed by patch clamp technique.The difference values(Vts-Vr_(1-4))of resting membrane potential(Vr_(1-4))and threshold potential(Vts_(1-4))of action potential of 4 times,the absolute refractory period(ARP_(1-4))and the action potential interval(ISI_(1-4))in theγ-aminobutyric neurons of endo olfactory cortex of brain among three groups were compared.Results:The ARP_(1-4) of model group was significantly longer than that of control group and experimental group(t=3.551,t=4.052,t=3.854,t=4.202,P<0.05),respectively.There were significant differences in ARP_(1-4) among three groups(F=7.96,F=4.65,F=5.06,F=8.13,P<0.05).The Vts-Vr_(1-4) of experimental group was significantly shorter than that of model group and control group(t=4.215,t=4.551,t=3.965,t=4.023,P<0.05),respectively.The ISI_(1-4) of model group was significantly longer than that of control group(t=3.662,t=3.564,t=3.266,t=3.537,P<0.05),respectively.And the ISI_(1-4) of model group was significantly longer than that of control group and experimental group(F=8.84,F=6.22,F=11.28,F=13.42,P<0.05),respectively.Conclusion:Lev intervention can significantly shorten the Vts-Vr,ARP and ISI ofγ-aminobutyric neurons in endo olfactory cortex of epileptic mice,and can significant improve the coding ability and excitability ofγ-aminobutyric
