摘 要:为比较不同方式提取的蛇莓抽提物的活性差异,采用牛津杯法和微量肉汤二倍稀释法检测蛇莓抽提物的抑菌活性,以FRAP法、ABTS法、DPPH法、超氧阴离子自由基(O_(2)^(-)·)清除法评估抗氧化活性,通过测定NO和MDA含量、SOD活性、i NOSm RNA表达变化评价蛇莓多糖抗脂质过氧化作用。结果显示,蛇莓多糖无抑菌活性,蛇莓醇提物的抑菌效果为7/11,蛇莓水提物的抑菌效果为5/11,蛇莓醇提物和蛇莓水提物均对化脓隐秘杆菌的生长呈高度抑制,最小抑菌浓度为0.98 mg/m L;蛇莓多糖的ABTS和DPPH自由基清除能力与Vc间无显著差异性(P>0.05),高于蛇莓醇提物和水提物差异极显著(P<0.01);蛇莓多糖能抑制LPS所诱导的RAW264.7细胞中产生NO和MDA、i NOS m RNA的表达(P<0.01),显著增强总SOD活力(P<0.01)。结果提示,蛇莓多糖虽然没有其醇提物和水提物的显著抑菌活性,但其抗氧化活性优于蛇莓醇提物和水提物,且能降低LPS诱导的巨噬细胞脂质过氧化反应损伤,蛇莓提取物有防治畜禽细菌感染性疾病前景。In order to compare the activity differences of extracts from Duchesnea indica(Andr.)Focke by different extraction methods,Oxford cup method and double dilution method of microbroth were used to detect the antibacterial activity of the extracts.FRAP method,ABTS method,DPPH method and superoxide free radical(O_(2)^(-)·)scavenging method were used to evaluate the antioxidant activity.The contents of NO and MDA,SOD activity and m RNA expression of i NOS were determined to evaluate the anti-lipid peroxidation effect of D.indica polysaccharide.The results showed that the D.indica polysaccharide had no antimicrobial activity.The antimicrobial effect of the alcohol extract of D.indica was 7/11,and the antimicrobial effect of the aqueous extract of D.indica was 5/11,and both extracts had high inhibition on the growth of Trueperella pyogenes and the minimum inhibition concentration was 0.98 mg/m L.The ABTS and DPPH free radical scavenging abilities of D.indica polysaccharide were not significantly different than that of Vc(P>0.05),and were higher than that of D.indica alcohol extract and aqueous extract,and the difference was very significant(P<0.01).D.indica polysaccharide inhibited significantly the generation of NO and MDA contents and expression of i NOS m RNA in RAW264.7 cells induced by LPS(P<0.01),and the total SOD activity in RAW264.7 cells was increased significantly(P<0.01).The results indicated that the polysaccharides from D.indica had better antioxidant activity than the alcohol and aqueous extracts of D.indica,and it can alleviate LPS-induced lipid peroxidation in macrophages.D.indica extracts had a promising prospect in the prevention and treatment of bacterial infectious diseases of livestock and poultry.
