摘 要:【目的】为明确马铃薯NAC转录因子在响应铁盐(Fe^(2+))胁迫时的表达特性,本试验对铁盐处理下马铃薯NAC转录因子相对表达量的时空差异进行分析。【方法】基于实验室前期对苗龄20 d的马铃薯品种东农312组培苗施加缺铁(1μmol/L)胁迫处理0、48 h,对其进行转录组测序,发现7个差异表达的NAC转录因子。本试验对MS培养基设置3个Fe^(2+)梯度(缺铁1μmol/L、对照100μmol/L和过量铁500μmol/L),分别用于处理组培条件下继代生长20 d的东农312脱毒试管苗,处理时间分别为0、48和96 h,应用qRT-PCR技术分析上述7个NAC转录因子在根、茎、叶中的相对表达量;并对NAC1~NAC7编码蛋白进行生物信息学分析。【结果】NAC1~NAC7在根、茎中均受缺铁、过量铁处理诱导表达,转录表达水平在处理48、96 h时较0 h时变化显著。在叶中,NAC1~NAC7在这两个时间点的表达量较0 h无显著上调。NAC1~NAC7编码蛋白含有192~413个氨基酸,均含有NAM结构域,预测分子量为22.81~46.98(kDa),PI值为4.65~9.23。【结论】马铃薯NAC转录因子参与响应铁盐胁迫。在缺铁和过量铁胁迫下,7个NAC基因在根和茎中均呈现显著差异表达,在叶中无显著上调表达。【Objective】To determine the expression characteristics of potato NAC transcription factor in response to iron salt(Fe^(2+))stress,the temporal and spatial differences of relative expression levels of potato NAC transcription factor under iron salt treatment were analyzed in this experiment.【Method】Based on the early stage of the laboratory,the tissue cultured seedlings of the 20-day-old potato variety Dongnong 312 were treated with iron deficiency(1μmol/L)for 0 and 48 hours,the transcriptome was sequenced,and 7 differentially expressed NAC transcription factors were found.In this experiment,three Fe^(2+)gradients(iron deficiency 1μmol/L,control 100μmol/L,and excess iron 500μmol/L)were set up on MS medium,and they were used to treat subculture growth under tissue culture conditions.The 20-day Dongnong 312 virus-free test-tube seedlings were treated at 0,48 and 96 hours,respectively.The relative expression levels of the above 7 NAC transcription factors in roots,stems and leaves were analyzed by qRT-PCR technology,and bioinformatics analysis of NAC1-NAC7 encoded protein was conducted.【Result】The expression of NAC1-NAC7 in roots and stems was induced by iron deficiency and excessive iron treatments,and the transcription expression level changed significantly at 48 and 96 hours compared with 0 hour.In leaves,the expression of NAC1-NAC7 at these two time points was not significantly up-regulated compared with 0 hour.The NAC1-NAC7 encoded protein contained 192-413 amino acids,all of which contained NAM domains.The predicted molecular weight was 22.81-46.98(kDa),and the PI value was 4.65-9.23.【Conclusion】Potato NAC transcription factors participate in response to iron salt stress.Under the stress of iron deficiency and excessive iron,the seven NAC genes showed significant differential expression in roots and stems,and no significant up-regulation expression in leaves.
