摘 要:目的:优选白芷挥发油最佳提取工艺,并对其抗氧化活性进行研究。方法:采用Box-Behnken设计响应面法,考察液料比、浸泡时间、提取时间及粉碎粒度4个因素对白芷挥发油得率的影响,分别采用多元线性方程及二项式方程拟合实验结果,确定优化组合条件。结果:最佳提取工艺为:粉碎粒度为65目,液料比为20∶1(mL/g),浸泡时间为2.3h,提取时间为5.4h,在此条件下挥发油得率为2.273%。对羟自由基的IC50为0.882mg/mL,对DDPH自由基的IC50为0.722mg/mL。结论:优化后的白芷挥发油提取工艺简便、稳定、可行、所得挥发油具有较好的抗氧化作用,此研究可为白芷挥发油抗氧化产品的综合开发提供理论依据,也为其他植物挥发油的提取和抗氧化性研究提供参考。Objective:To optimize extraction technology of volatile oil from Angelica dahurica and study its antioxidant activity.Methods:Response surface methodology was used to investigate the effects of liquid-solid ratio,soaking time,extraction time and particle size on the yield of volatile oil from Angelica dahurica.The optimal extraction conditions were determined by multivariate linear equation and binomial equation.Results:The optimal extraction conditions were:particle size 65 mesh,liquid-solid ratio 20∶1(mL/g),soaking time 2.3 h and extraction time of 5.4 h.Under the above conditions,the yield of volatile oil was 2.273%.The IC50 values for hydroxyl radical and DDPH radical were 0.882 mg/mL and 0.722 mg/mL respectively.Conclusion:The optimized extraction process is simple,stable and feasible.The extracted volatile oil has good antioxidant effect.The study provides a theoretical basis for comprehensive development of antioxidant products from Angelica dahurica volatile oil.The study also provides a reference for extraction and antioxidant study of volatile oil from other plants.
