摘　　要：目的研究GATA5对肾癌HEK-293细胞系增殖调控及对细胞重编程因子EpCAM表达的影响。方法构建pTT5-GATA5表达载体并进行基因序列测序验证;四甲基偶氮唑盐(MTT)法检测细胞增殖率;细胞划痕实验检测细胞划痕修复情况;实时定量反转录-聚合酶链反应(RT-PCR)检测基因mRNA表达;蛋白免疫印迹(Western Blot)检测蛋白表达。结果GATA5抑制HEK-293细胞增殖;GATA5抑制HEK-293细胞划痕修复;GATA5抑制EpCAM的mRNA和蛋白表达。结论GATA5可以抑制肾癌细胞HEK-293细胞的增殖率并抑制EpCAM表达。Objective:To study the effect of GATA5 on the proliferation regulation of renal cell carcinoma HEK-293 cell line and the expression of cell reprogramming factor EpCAM.Methods:Construction of pTT5-GATA5 expression vector and sequence verification of gene sequence;MTT method to detect cell proliferation rate;cell scratch test to detect cell scratch repair;RT-PCR to detect gene mRNA expression;western blot detect protein expression.Results:GATA5 inhibits HEK-293 cell proliferation;GATA5 inhibits HEK-293 cell scratch repair;GATA5 inhibits EpCAM mRNA and protein expression.Conclusion:GATA5 can inhibit the proliferation rate of renal cancer cell HEK-293 cells and inhibit the expression of EpCAM.
