摘 要:目的研究胰高血糖素样肽-2(glucagon like peptide-2,GLP-2)基于磷脂酰肌醇3-激酶(phosphoionsitide 3-kinase,PI3K)/蛋白激酶B(protein kinase B,Akt)信号通路对急性胰腺炎模型的干预作用。方法选取30只Wistar雄性大鼠,选取10只分为正常组,其余20只建立急性胰腺炎模型,成功18只,随机分为模型组和GLP-2试剂组(0.25 mg/kg),每组9只。检测血清淀粉酶、脂肪酶、二胺氧化酶(diamine oxidase,DAO)、D-乳酸、白细胞计数、C反应蛋(c-reactive protein,CRP)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)水平,统计胰腺干湿重比、病理学观察及胰腺病理评分,检测PI3K、Akt、p-Akt蛋白表达。结果模型组和GLP-2试剂组血清淀粉酶、脂肪酶、DAO、D-乳酸、白细胞计数、CRP、TNF-α水平,胰腺干湿重比、胰腺病理评分升高,2组差异有统计学意义(P<0.05)。GLP-2试剂组血清淀粉酶、脂肪酶、DAO、D-乳酸、白细胞计数、CRP、TNF-α水平,胰腺干湿重比、胰腺病理评分低于模型组,差异有统计学意义(P<0.05)。模型组和GLP-2试剂组PI3K、Akt、p-Akt蛋白表达降低;GLP-2试剂组PI3K、Akt、p-Akt蛋白表达高于模型组,差异有统计学意义(P<0.05)。结论GLP-2试剂通过调控PI3K/AKT信号通路相关蛋白,对急性胰腺炎模型干预效果显著,能改善模型胰腺组织以及肠道黏膜损伤状况,抑制机体炎性反应。Objective To investigate the intervention effects of glucagon like peptide-2(GLP-2)based on the signal pathway of phosphoinositide 3-kinase(PI3K)/protein kinase B(Akt)in patients with acute pancreatitis.Methods Thirty male Wistar rats were enrolled in the study,of whom,10 rats were enroled as control group,and the animal models with acute pancreatitis were established in the other 20 rats,moreover,the rat models were established successfully in 18 rats,who were randomly divided into model group and GLP-2 reagent group(0.25mg/kg),with 9 rats in each group.The serum levels of amylase,lipase,diamine oxidase(DAO),D-lactate,leukocyte count,C-reactive protein(CRP),tumor necrosis factor-α(TNF-α)were detected.Moreover the ratio of dry to wet weight of pancreas,pathological observation and pancreatic pathological score were performed.In addition expression levels of PI3K,Akt and p-Akt protein were detected.Results The levels of serum amylase,lipase,DAO,D-lactic acid,WBC count,CRP,TNF-αlevels,dry-wet ratio of pancreas,and pancreatic pathology score in model group and the GLP-2 group were significantly increased(P<0.05).The levels of serum amylase,lipase,DAO,D-lactic acid,WBC count,CRP and TNF in GLP-2 reagent group were significantly lower than those in model group(P<0.05).The expression levels of PI3K,Akt and P-Akt proteins in model group and GLP-2 reagent group were decreased,which in GLP-2 reagent group were significantly higher than those in model group(P<0.05).Conclusion GLP-2 can regulate the PI3K/Akt signal pathway related protein,which has significant intervention effects on the rat models with acute pancreatitis,which can improve the damage of pancreatic tissue and intestinal mucosa,and inhibit the inflammatory response of the body.
