摘 要:目的探讨益心泰有效组分对慢性心衰兔心肌组织CaN的影响。方法将造模成功的慢性心衰兔分为HF模型组、益心泰有效组分低、中、高剂量组和氯沙坦钾组,另设假手术组;各用药组给予相应药物灌胃,HF模型组和假手术组灌胃同等体积的生理盐水,1次/天,共干预4周。实验结束后比较各组兔心肌组织形态学,心功能指标,心肌细胞内Ca^(2+)浓度,心肌组织CaN mRNA与蛋白表达及其活性。结果益心泰有效组分能减轻慢性心衰兔的心肌损伤,提高左心室射血分数(LVEF)、左心室短轴缩短率(LVFS)、E峰与A峰的比值(E/A)(P<0.01),下调心肌细胞内Ca^(2+)浓度,下调心肌组织CaN活性、CaNmRNA及蛋白表达(P<0.01)。结论益心泰有效组分可抑制慢性心衰兔心肌组织CaN活性、CaN mRNA及蛋白表达,改善心肌重构,提高心功能。Aim To explore the effects of the effective components of Yi-xin-tai on the CaN in myocardial tissues of rabbits with chronic heart failure(CHF).Methods The rabbits successfully modeled were divided into high,middle,and low dose groups of the effective components of Yi-xin-tai,losartan potassium group and HF model group.Besides,a sham operation group was set up.Each treatment group was given corresponding drugs by gavage.And equal volume of physiological saline was administered to rabbits of HF model group and sham operation group by gastrogavage.The intervention lasted for four weeks,once per day.After treatment,the morphology of myocardial tissues,cardiac function indicators,the concentration of[Ca^(2+)]i in myocardial cells,CaN activity,mRNA and protein expression in myocardial tissues were observed.Results The effective components of Yi-xin-tai reduced the myocardial injury in chronic heart failure,and increased the left ventricular ejection fraction(LVEF),the left ventricular short axis reduced rates(LVFS)and the ratio of E/A ratio(P<0.01).Besides,it decreased the concentration of[Ca^(2+)]i in myocardial cells,and decreased the CaN activity,mRNA and protein expression in myocardial tissues(P<0.01).Conclusions The effective components of Yi-xin-tai can inhibit CaN activity,mRNA and protein expression in myocardial tissues of CHF rabbits,improve myocardial remodeling,and improve heart function.
