摘 要:目的通过观察系统性硬皮病(systemic scleroderma,SSc)小鼠经刺山柑总生物碱治疗后,其Wnt通路中Wnt2、Wnt3a及β-catenin的表达水平,探讨其对SSc Wnt通路的调控作用。方法BALB/c小鼠皮下注射博来霉素复制SSc模型。给予不同量刺山柑总生物碱,以及阳性药青霉胺干预。取皮肤,Western blot测Wnt3a、β-catenin蛋白表达,q-RT PCR测Wnt2 mRNA表达,免疫组化检测Wnt3a蛋白表达。结果刺山柑总生物碱中、高剂量组可明显降低SSc小鼠皮肤β-catenin的表达水平(P<0.05),各剂量药物组可明显降低Wnt2、Wnt3a蛋白表达(P<0.01)。结论刺山柑总生物碱可降低SSc小鼠皮肤Wnt2、Wnt3a和β-catenin表达,对其过表达的Wnt通路有一定抑制作用。Aim To observe the expression of Wnt2,Wnt3a andβ-catenin in the Wnt pathway of systemic scleroderma(SSc)mice treated with total alkaloids of Capparis spinosa,and to explore the regulation of Wnt2,Wnt3a andβ-catenin on SSC Wnt pathway.Methods BALB/c mice were injected with bleomycin to establish SSC model.Different doses of total alkaloids from Capparis spinosa and penicillamine were administered.The expressions of Wnt3a andβ-catenin were detected by Western blot,Wnt2 mRNA expression by q-RT PCR and Wnt3a protein expression by immunohistochemistry.Results The expression level ofβ-catenin in skin of SSC mice significantly decreased in the medium and high dose groups(P<0.05),and the protein expressions of Wnt2 and Wnt3a significantly decreased in all doses of total alkaloids of Capparis spinosa(P<0.05 or P<0.01).Conclusions The total alkaloids of Capparis spinosa can reduce the expression of Wnt2,Wnt3a andβ-catenin in SSc mice,and inhibit the overexpression of Wnt pathway in SSc mice.
