摘 要:目的探讨阳和汤对Lewis肺癌荷瘤小鼠的抑瘤效应及对肿瘤免疫微环境的影响。方法将50只雄性C57BL/6小鼠右侧前腋皮下接种浓度为2×107/ml的Lewis肺癌细胞悬液0.2 ml以建立Lewis肺癌小鼠模型,造模成功后,按照随机数字表法分为荷瘤模型组、环磷酰胺组、阳和汤高剂量组、阳和汤中剂量组和阳和汤低剂量组,每组10只,分别给予生理盐水、25 mg/kg环磷酰胺和27.5、18.3、9.2 g/kg的阳和汤连续灌胃10 d,1次/d。观察各组小鼠的体重和瘤重,分别用免疫组织化学法和免疫荧光技术检测肿瘤组织中白细胞介素(IL)-2、转化生长因子(TGF)-β和细胞毒性T淋巴细胞(CTL)和调节性T细胞(Treg)标志物CD3^(+)CD8^(+)、CD4^(+)Foxp3^(+)的表达。结果与荷瘤模型组比较,阳和汤高、中剂量组小鼠体重增加(P<0.05或P<0.01);与环磷酰胺组比较,阳和汤高、中、低剂量组小鼠体重增加(P<0.01);与阳和汤高剂量组比较,阳和汤低剂量组小鼠体重降低(P<0.05)。与荷瘤模型组比较,阳和汤高剂量组小鼠瘤重降低(P<0.05);与环磷酰胺组比较,阳和汤中、低剂量组小鼠瘤重增加(P<0.05)。抑瘤率由高到底分别为环磷酰胺组、阳和汤高剂量组、阳和汤中剂量组、阳和汤低剂量组。与荷瘤模型组比较,阳和汤高、中、低剂量组小鼠Lewis肺癌组织IL-2表达增加(P<0.05或P<0.01);与环磷酰胺组比较,阳和汤高、中、低剂量组小鼠Lewis肺癌组织IL-2表达增加(P<0.01);与阳和汤高剂量组比较,阳和汤中、低剂量组小鼠Lewis肺癌组织IL-2表达降低(P<0.01)。与荷瘤模型组比较,阳和汤高、中剂量组小鼠Lewis肺癌组织TGF-β表达降低(P<0.01);与环磷酰胺组比较,阳和汤中、低剂量组小鼠Lewis肺癌组织TGF-β表达增加(P<0.05)。与荷瘤模型组比较,阳和汤高、中、低剂量组小鼠Lewis肺癌组织CD3^(+)CD8^(+)表达增加(P<0.05或P<0.01);与环磷酰胺组比较,阳和汤高、中、低剂量组小鼠LewiObjective To investigate the antitumor effect and the effect of tumor immune microenvironment of Yanghe Decoction on Lewis lung cancer mice. Methods A total of 50 male C57BL/6 mice were subcutaneously inoculated with 0.2 ml Lewis lung cancer cell suspension at the concentration of 2×107/ml in the right anterior axilla to establish Lewis lung cancer mouse model. After the model was made, according to random number table method, these mice were divided into tumor-bearing model group, Cyclophosphamide group, Yanghe Decoction high concentration group, Yanghe Decoction medium concentration group, and low concentration group, with 10 mice in each group. Normal saline, 25 mg/kg Cyclophosphamide, 27.5, 18.3, and 9.2 g/kg Yanghe Decoction were given by intragastric administration for 10 days, respectively, one time per day. The body weight and tumor weight of mice in each group were observed. The expressions of interleukin (IL)-2, transforming growth factor (TGF)-β, and cytotoxic lymphocyte (CTL) and regulatory T cell (Treg) markers CD3^(+)CD8^(+) CD4^(+)Foxp3^(+) in tumor tissues were detected by immunohistochemistry and immunofluorescence, respectively. Results Compared with tumor-bearing model group, the body weight of mice in Yanghe Decoction high, medium concentration groups were increased (P < 0.05 or P < 0.01);compared with Cyclophosphamide group, the body weight of mice in Yanghe decoction high, medium, and low concentration groups were increased (P < 0.01);and compared with Yanghe Decoction high concentration group, the body weight of mice in Yanghe Decoction low concentration group was decreased (P < 0.05). Compared with tumor-bearing model group, tumor weight of mice in Yanghe Decoction high concentration group was decreased (P < 0.05);and compared with Cyclophosphamide group, tumor weight of mice in Yanghe Decoction medium, low concentration groups were increased (P < 0.05).
