摘  要:目的通过腹腔注射戊四氮(PTZ)构建大鼠癫痫模型,分析葱白提取物对模型大鼠脑电图(EEG)、学习记忆能力、半胱氨酸天冬氨酸蛋白酶(Caspase)-3和抗凋亡因子(Livin)的影响,探讨葱白提取物神经保护效果及作用机制。方法取购于哈尔滨医科大学实验动物中心的雄性的健壮成年大鼠30只,随机分为正常组(生理盐水腹腔注射+生理盐水灌胃)、模型组(PTZ腹腔注射+生理盐水灌胃)、治疗组(PTZ腹腔注射+葱白提取物灌胃)各10只。3组完成前述操作周期后描记EEG、监测学习记忆能力、记录凋亡蛋白Caspase-3、Livin的改变规律和特点。结果经葱白提取物灌胃治疗后,模型组EEG中的尖波或者棘波有所恢复,变成了平缓的慢波和小波;且学习记忆能力得到了显著改善。模型组皮质及海马Caspase-3表达的阳性细胞数目显著变多,而海马区阳性细胞数又明显多于颞叶皮质区。治疗组Livin在皮质和海马区表达水平显著上调(P<0.05)。结论葱白提取物可通过上调Livin阳性细胞数、下调Caspase-3阳性细胞数来削弱并抑制神经细胞的坏死和凋亡,加快大脑局部的血液循环来增强学习记忆能力;改善EEG波形来达到神经保护作用。Objective To investigate the protective effect and mechanism of onion extract on nerve.Methods The rat model of epilepsy was established by intraperitoneal injection of pentatetrazol(PTZ).Thirty healthy adult male rats purchased from Harbin Medical University were selected into normal group(intraperitoneal injection of normal saline+gavage of normal saline),model group(intraperitoneal injection of PTZ+gavage of normal saline),and treatment group(intraperitoneal injection of PTZ+gavage of scallion extract)with 10 rats in each group.Electroencephalography(EEG),learning and memory ability,and the changes and characteristics of apoptotic proteins caspase-3 and Livin were recorded for all the rats in the three groups.Results After intragastric treatment with scallion extract,the sharp wave or spike wave in EEG of rats in epilepsy model group recovered to gentle slow wave and wavelet wave.And the ability to learn and remember had been significantly improved.The number of Caspase-3 positive cells in cortex and hippocampus increased significantly in model group,and the number of positive cells in hippocampus was significantly higher than that in temporal cortex.The expression of Livin was up-regulated in cortex and hippocampus of the treatment group than that of model group(P<0.05).Conclusions Scallion extract could weaken and inhibit the necrosis and apoptosis of nerve cells by up-regulating the number of Livin positive cells and down-regulating the number of Caspase-3 positive cells,and accelerate the local blood circulation in the brain to enhance learning and memory ability,improve EEG wave form to achieve neuroprotective effects.
