摘  要:目的:研究受体相互作用蛋白激酶1(RIP1)在小鼠实验性牙周炎模型中的作用和可能机制。方法:提取牙周炎患者及牙周健康者的牙龈组织,Western blot检测RIP1蛋白的表达。在RIP1条件性基因敲除(RIP1^(-/-))小鼠和C57野生型小鼠上颌第二磨牙建立牙周炎模型,用Micro-CT扫描成像分析对比两组小鼠实验侧牙槽骨丧失量。提取2组小鼠对照侧及实验侧牙龈组织,用实时荧光定量聚合酶链反应检测相关炎症因子mRNA的表达差异。结果:牙周炎患者牙龈组织RIP1蛋白的表达降低(P<0.01)。建立牙周炎模型后,RIP1^(-/-)小鼠牙槽骨丧失量增加(P<0.01),牙龈组织中多个促炎和抗炎因子表达改变(P<0.05)。结论:RIP1可能通过调节炎症因子的表达减轻实验性牙周炎小鼠的牙槽骨吸收。Objective:To study the role and possible mechanism of receptor interaction protein kinase 1(RIP1)in experimental periodontitis models of mice.Methods:The gingival tissue was obtained from patients with periodontitis and periodontal health respectively,the expression of RIP1 protein was detected by Western blot.Periodontitis models were established at the cervical area of second maxillary molar of RIP1-conditional knock-out(RIP1^(-/-))mice and C57 wild-type(WT)mice,the amount of experimental lateral alveolar bone loss in the 2 groups was compared using Micro-CT scanning imaging analysis.The mRNA expression of 6 inflammatory-related factors in the periodontal tissue of both healthy and affected gingival tissues of RIP1^(-/-) and WT mice was detected by qRT-PCR.Results:Lower expression level of RIP1 protein was observed in periodontitis group compared with that in healthy control group(P<0.01).Compared with WT mice in the loss of alveolar bone increased in RIP1^(-/-) mice periodontitis model(P<0.01),the expression of 6 pro-inflammatory and anti-inflammatory factors in gingival tissue of RIP1^(-/-) mice was statistically changed(P<0.05).Conclusion:RIP1 may reduce alveolar bone resorption in experimental periodontitis mice by regulating the expression of inflammatory factors.
