摘　　要：目的:探讨CXC类趋化因子配体2(CXCL2)通过募集肿瘤相关中性粒细胞(TANs)促进肿瘤微环境(TME)中舌癌细胞TSCCA侵袭能力的研究。方法:实验建立牙龈卟啉单胞菌(P.gingivalis)感染TSCCA细胞的共培养模型,利用细胞趋化实验检测共培养组对TANs的趋化能力;细胞划痕实验、Transwell侵袭实验、CCK-8增殖实验和qRT-PCR实验检测实验组TSCCA+P.gingivalis+TANs与对照组TSCCA+P.gingivalis+TANs+SCH527123,TSCCA、TSCCA+P.gingivalis、TSCCA+TANs组中细胞迁移、侵袭、增殖的能力,比较差异。结果:实验组相比对照组细胞趋化、迁移、侵袭、增殖能力明显升高(P<0.001)。结论:P.gingivalis感染TSCCA的口腔TME中,CXCL2通过募集TANs促进TSCCA迁移和侵袭能力。Objective:To investigate the effects of CXC chemokine ligands(CXCL2)on the invasion of tongue squamous cell carcinoma(TSCCA)cells in tumor microenvironment(TME)by recruiting tumor-associated neutral granular cells(TANs).Methods:The co-culture model of P.gingivalis infection with TSCCA cells was established,The chemotactic ability of the TSCCA cells of co-culture group to TANS was detected by chemotactic assay.Cell scratch assay,Transwell invasion assay,CCK-8 proliferation assay and qRT-PCR assay were used to detect the ability of cell migration,invasion and proliferation in the experimental group of TSCCA+P.gingivalis+TANs and control groups of TSCCA+P.gingivalis+TANs+SCH527123,TSCCA,TSCCA+P.gingivalis and TSCCA+TANs.The differences were compared.Results:Compared with the control groups,the chemotaxis,migration,invasion,proliferation ability in the experimental group were significantly increased(P<0.001).Conclusion:CXCL2 promotes the invasion of TSCCA cells by recruiting TANs in oral TME infected with P.gingivalis.
