摘 要:目的探讨双歧杆菌-单纯疱疹病毒胸苷激酶基因-丙氧鸟苷(BI-HSV-TK/GCV)系统对裸小鼠胃癌细胞增殖的抑制效应和分子机制。方法建立裸小鼠胃癌实验模型,检测裸小鼠的体重、肿瘤体积。运用TUNEL法观察裸小鼠胃癌细胞的凋亡情况,运用Western blot实验、RT-PCR实验分别测定胃癌组织中Caspase 3、Caspase 8、Fas及FAP-1蛋白及其mRNA表达情况。结果BI-HSV-TK/GCV系统能够改善裸小鼠的体重,有效抑制其胃癌肿瘤的体积,促进胃癌细胞的凋亡,诱导胃癌细胞凋亡相关基因Caspase 3、Caspase 8、Fas蛋白以及mRNA表达水平增加,FAP-1蛋白以及mRNA表达水平降低。结论BI-HSV-TK/GCV系统能够有效抑制胃癌细胞在裸小鼠体内成瘤的体积,并促进胃癌细胞的凋亡。Objective To investigate the inhibitory effect and molecular mechanism of Bifidobacterium-herpes simplex virus thymidine kinase gene-ganciclovir(BI-HSV-TK/GCV)system on the gastric cancer cells proliferation in nude mice.Methods The experimental model of gastric cancer in nude mice was established,and the body weight and tumor volume of nude mice were measured.The apoptosis of gastric cancer cells in nude mice was observed by TUNEL method,and the expressions levels of Caspase 3,Caspase 8,Fas and FAP-1 proteins and their mRNA in gastric cancer tissues were measured by Western blot test and RT-PCR test.Results BI-HSV-TK/GCV system could improve the body weight of nude mice,effectively inhibit the volume of gastric cancer tumor,promote the apoptosis of gastric cancer cells,induce the increase of protein and their mRNAs expressions level of apoptosis related gene such as Caspase 3,Caspase 8,Fas,and the decrease of expression level of FAP1 protein and its mRNA.Conclusion BI-HSV-TK/GCV system can effectively inhibit the tumor formation volume of gastric cancer cells in nude mice and promote the apoptosis of gastric cancer cells.
