甲状腺乳头状癌组织中miR-182表达情况及其与临床病理特征的关系

摘　　要：目的探讨甲状腺乳头状癌组织中miR-182表达情况及其与临床病理特征的关系。方法收集经病理学检测确诊的甲状腺乳头状癌组织98例和相应的癌旁组织98例(对照组),采用定量聚合酶链反应(quantitative polymerase chain reaction,qPCR)测定甲状腺乳头状癌组织以及癌旁组织中miR-182水平,分析其与患者临床病理特征以及预后的关系。结果甲状腺乳头状癌组miR-182表达水平高于对照组[(1.89±0.48)vs(0.66±0.21),P<0.01]。病理学分期高、TMN分期高、浸润深度深、有淋巴血管间隙浸润和有淋巴结转移的甲状腺乳头状癌患者miR182阳性表达率高(均P<0.01)。甲状腺乳头状癌组中,以miR-182水平均值(1.89)为判定点,miR-182水平≥1.89的患者3年生存率低于miR-182水平<1.89的患者(27.9%vs 80.0%,P<0.01)。结论甲状腺乳头状癌中miR-182表达升高,miR-182在甲状腺乳头状癌的发生和发展过程中起促进作用。miR-182低表达的甲状腺乳头状癌患者能获得较好的预后。Objective To investigate the expression of miR-182 in papillary thyroid carcinoma and its relationship with clinicopatho-logical features.Methods Ninety-eight cases of papillary thyroid carcinoma tissues confirmed by pathology(study group)and their cor-responding paracancerous tissues(control group)were collected,and quantitative polymerase chain reaction(qPCR)was used to determine the level of miR-182 in thyroid papillary carcinoma tissues and paracancerous tissues.The relationship of patients'clinicopathological characteristics and prognosis was analyzed.Results The expression of miR-182 in the study group was higher than that in the control group[(1.89±0.48)vs(0.66±0.21),P<0.01].The expression of miR-182 was positively correlated with pathological grade,TMN stage,depth of invasion,lymphatic vascular infiltration,and lymph node metastasis(all P<0.01).In the study group,taking the mean miR-182 level of 1.89 as the decision point,the 3-year survival rate of the miR-182 positive(≥1.89)group was significantly lower than that of the miR-182 negative(<1.89)group(27.9%vs 80.0%,P<0.01).Conclusions The expression of miR-182 is elevated in papillary thyroid carcinoma,and miR-182 may play a role in promoting the development of papillary thyroid carcinoma.Patients with low expression of miR-182 may have better prognosis.
