摘　　要：为了研究克氏原螯虾LC3基因的序列及其生物信息学,分离并提取肝胰腺组织,并克隆了其LC3基因ORF全长序列。结果表明,克氏原螯虾LC3基因ORF序列片段为369 bp,编码122个氨基酸。结构分析显示,LC3基因存在2个结构域,证明了LC3基因在细胞自噬中存在多种功能。序列比对及系统进化树分析表明,克氏原螯虾与节肢动物进化关系最接近,其LC3基因序列具有节肢动物的典型特征,在进化过程中保守性较高。荧光定量PCR显示,克氏原螯虾LC3基因在肝胰腺中表达水平最高,与肠道相比有显著差异(P<0.05),与心脏和肌肉无显著差异(P>0.05);心脏和肌肉中LC3基因的表达量次之,但与肠道相比差异显著(P<0.05);肠道中LC3表达量最低。该研究结果可为深入研究LC3基因表达与动物营养调控提供基础依据和参考。In order to study the sequence of LC3 gene and its bioinformatics of Procambarus clarkii,hepatopancreas tissue was isolated and extracted,the full-length ORF sequence of LC3 gene was cloned.The results showed that the ORF sequence fragment of the LC3 gene of P.clarkii was 369 bp,encoding 122 amino acids.Structural analysis showed that the LC3 gene had two domains,proving that the LC3 gene had multiple functions in autophagy.Sequence alignment and phylogenetic tree analysis showed that P.clarkii had the closest evolutionary with arthropods,and its LC3 gene sequence had typical arthropod characteristics and it was highly conserved during evolution.Fluorescence quantitative PCR showed that P.clarkii LC3 gene was expressed at the highest level in the hepatopancreas,and it was significantly different from the intestine(P<0.05),but it was not significantly different from the heart and muscle(P>0.05);the muscle expression was the second,but it was still significantly different from the intestine(P<0.05);the expression of LC3 in the intestine was the lowest.The research results could provide basic basis and reference for in-depth study on LC3 gene expression and animal nutrition regulation.
