摘 要:目的探究LncRNA-SNHG3(SNHG3)对乳腺癌细胞增殖的影响及其可能的作用机制。方法慢病毒包装SNHG3干扰质粒和过表达质粒,慢病毒液感染乳腺癌细胞,获得干扰SNHG3(sh-SNHG3)和过表达SNHG3(oe-SNHG3)的稳转细胞系,RT-qPCR检测SNHG3 mRNA表达;CCK-8检测细胞活力;克隆形成实验检测细胞增殖;Western blot法检测Gli1蛋白表达。结果与sh-NC组比较,sh-SNHG3组细胞中SNHG3 mRNA的表达明显降低(P<0.05),12、24、48和72h细胞活力、克隆形成数和Gli1蛋白表达明显降低(P<0.05);与oe-NC组比较,oe-SNHG3组细胞中SNHG3 mRNA的表达明显升高(P<0.05),12、24、48和72h细胞活力、克隆形成数和Gli1蛋白表达明显升高(P<0.05)。与oe-SNHG3组比较,oe-SNHG3+cyclopamine组细胞中SNHG3 mRNA表达差异无统计学意义(P>0.05),12、24、48和72h细胞活力、克隆形成数和Gli1蛋白表达明显降低(P<0.05)。结论SNHG3通过激活Hedgehog通路促进乳腺癌细胞增殖。Objective To investigate the effect of LncRNA-SNHG3(SNHG3)on the proliferation of breast cancer cells and its possible mechanism.Methods Lentivirus was used to package SNHG3 interference plasmid and overexpression plasmid.Stable cell lines were obtained by infecting breast cancer cells with lentivirus to interfere with SNHG3(sh-SNHG3)and overexpress SNHG3(oe-SNHG3),SNHG3 mRNA expression was detected by RT-qPCR,cell viability was detected by CCK-8,cell proliferation was detected by colony formation assay,and Gli1 protein expression was detected by Western blot.Results Compared with sh-NC group,the mRNA expression of SNHG3 in sh-SNHG3 group was significantly decreased(P<0.05),and the cell viability at 12,24,48 and 72h,number of colony formation and protein expression of Gli1 were significantly decreased(P<0.05).Compared with oe-NC group,the mRNA expression of SNHG3 in oe-SNHG3 group was significantly increased(P<0.05),and the protein expression of Gli1 was significantly decreased(P<0.05).Cell viability at 12,24,48 and 72h,number of colony formation and protein expression of Gli1 were significantly increased(P<0.05).Compared with the oe-SNHG3 group,the mRNA expression of SNHG3 in the oe-SNHG3+cyclopamine group was not significantly different(P>0.05),but the cell viability at 12,24,48 and 72h,number of colony formation and protein expression of Gli1 were significantly decreased(P<0.05).Conclusion SNHG3 promotes the proliferation of breast cancer cells by activating hedgehog pathway.
