摘  要:目的人参皂苷Rg1纳米颗粒(Rg1-NPs)已被证实能够主动靶向脑微血管内皮细胞(BMECs),文章进一步观察Rg1-NPs对缺血性脑卒中后血管新生的干预作用。方法制备耦联单克隆抗体OX26的纳米颗粒并用其包裹人参皂苷Rg1。在培养的BMECs细胞中分别加入PBS(对照组)、Rg1(Rg1组)和Rg1-NPs(Rg1-NPs组),采用MTT法、嵌套小室方法和小管形成实验分别检测Rg1-NPs对BMECs增殖、迁移和成管能力的影响。结果4℃储藏条件下人参皂苷Rg1泄露率为(5.98±0.58)%,在常温储藏条件下人参皂苷Rg1泄露率为(13.64±1.84)%。MTT实验结果显示,Rg1-NPs组(0.56±0.03)和Rg1组(0.49±0.02)BMECs细胞的增殖皆高于对照组(0.44±0.01),其中Rg1-NPs组BMECs细胞的增殖高于Rg1组(P<0.05)。嵌套小室方法检测的结果显示,Rg1-NPs组(55.33±5.03)和Rg1组(43.67±2.08)迁移的细胞数量均高于对照组(35.00±3.00),其中Rg1-NPs组迁移的细胞数量高于Rg1组,差异具有统计学意义(P<0.05)。小管形成实验结果显示,Rg1-NPs组和Rg1组管腔形成的网眼、点和管腔长度明显均高于对照组,其中Rg1-NPs组管腔形成的网眼、点和管腔长度高于Rg1组,差异均具有统计学意义(P<0.05)。结论Rg1-NPs能显著促进BMECs的增殖、迁移和成管能力,从而改善缺血性脑卒中后的血管新生。Objective Ginsenoside Rg1 nanoparticles(Rg1 NPs)have been proved to be able to actively target brain microvascular endothelial cells(BMECs).This paper further observed the intervention effect of Rg1 NPs on angiogenesis after ischemic stroke.Methods Nanoparticles coupled with monoclonal antibody OX26 were prepared and coated with ginsenoside Rg1.PBS(control group),Rg1(Rg1 group)and Rg1 NPs(Rg1 NPs group)were added to the cultured BMECs,and the effects of Rg1 NPs on the proliferation,migration and tubule formation of BMECs were detected by MTT method,nested chamber method and tubule formation experiment,respectively.Results The leakage rate of ginsenoside Rg1 was(5.98±0.58)%at 4℃and(13.64±1.84)%at room temperature.MTT assay results showed that the proliferation of BMECs cells in Rg1 NPs group(0.56±0.03)and Rg1 group(0.49±0.02)was higher than that in control group(0.44±0.01),and the proliferation of BMECs cells in Rg1 NPs group was higher than that in Rg1 group(P<0.05).The results of nested cell method showed that the number of migrating cells in Rg1 NPs group(55.33±5.03)and Rg1 group(43.67±2.08)was higher than that in control group(35.00±3.00),and the number of migrating cells in Rg1 NPs group was higher than that in Rg1 group,with statistical significance(P<0.05).The results of tubule formation experiment showed that the mesh,dots and lumen length in Rg1 NPs group and Rg1 group was significantly higher than those in the control group,and the mesh,dots and lumen length in Rg1 NPs group was higher than those in Rg1 group,with statistical significance(P<0.05).Conclusion Rg1 NPs can significantly promote the proliferation,migration and tubulogenesis of BMECs,thus improving angiogenesis after ischemic stroke.
