摘  要:目的:阐明活化的PPAR-γ调节溃疡性结肠炎(ulcerative colitis,UC)患者外周血Th1/Th2细胞分化的分子机制,为该药应用于UC的临床治疗提供新的实验依据。方法:将实验大鼠随机分为正常对照组、模型对照组(生理盐水灌胃)、SASP处理组(SASP,100 mg/kg)、低剂量罗格列酮组(罗格列酮2 mg/kg)、中剂量罗格列酮组(罗格列酮4 mg/kg)、高剂量罗格列酮组(罗格列酮8 mg/kg),每组10只。每日观察、记录各组小鼠的体重和大便情况,计算疾病活动指数(DAI)。于实验第10天取大鼠尾静脉血,以免疫组化方法检测血清中IFN-γ、IL-4水平表达。结果:与正常对照组比较,模型对照组与低剂量罗格列酮组DAI更高,差异有统计学意义(P<0.05);SASP处理组和中、高剂量罗格列酮组与正常对照组DAI比较差异无统计学意义(P>0.05)。模型对照组IFN-γ/IL-4高于正常对照组,差异有统计学意义(P<0.05),SASP处理组IFN-γ/IL-4与正常对照组比较差异无统计学意义(P>0.05);中、高剂量罗格列酮组IFN-γ/IL-4与正常对照组及SASP处理组比较差异无统计学意义(P>0.05)。结论:配体活化的PPAR-γ可调节UC患者外周血Th1/Th2细胞分化异常,从分子水平上阐明了其调控机制,为该药应用于UC的临床治疗提供新的实验依据。Objective:To elucidate the molecular mechanism of activated PPAR-γ in the differentiation of peripheral blood Th1/Th2 cells in patients with UC,and provide a new experimental basis for the clinical application of this drug in UC.Method:Rats were randomly divided into normal control group,model control group (normal saline intragastric administration),SASP treatment group (SASP,100 mg/kg),low-dose Rosiglitazone group (Rosiglitazone 2 mg/kg),medium-dose Rosiglitazone group (Rosiglitazone 4 mg/kg),high-dose Rosiglitazone group (Rosiglitazone 8 mg/kg),10 rats in each group.The body weight and stool of each group were observed and recorded daily,and the disease activity index (DAI) was calculated.The tail vein blood was taken from the 10th day of the experiment,and the expression of IFN-γand IL-4 in serum was detected by immunohistochemistry.Result:DAI was higher in the model control group and the low-dose Rosiglitazone group than in the normal control group,and the differences were statistically significant (P<0.05).There was no significant difference in DAI between the SASP treatment group,the medium,high dose Rosiglitazone group and the normal control group (P>0.05).The IFN-γ/IL-4 in model control group was significantly higher than that in normal control group (P<0.05),there was no significant difference in IFN-γ/IL-4 between SASP treatment group and normal control group (P>0.05);there was no significant difference in IFN-γ/IL-4 in the medium,high dose rosiglitazone group compared with the normal control group and SASP treatment group (P>0.05).Conclusion:Ligand-activated PPAR-γcan regulate the abnormal differentiation of Th1/Th2 cells in peripheral blood of patients with UC.The regulation mechanism is elucidated at the molecular level,which provides a new experimental basis for the clinical application of this drug in UC.
